In this paper with mountains of Rhodiola rosea male plant anther as explant, studies the different development period of pollen effects on anther callus induction;" Studies the different anther inoculation density effects on anther callus induction; To study the different kinds of basic culture medium, different kinds of plant hormones on the influence of anther callus induction and differentiation; To study the different light and temperature conditions on the anther callus induction and differentiation. The results showed that:1. Flower stalk length of anther callus induction had no obvious effect, flower diameter size of alpine Rhodiola anther callus induction, flower stalk length of anther callus induction had no obvious effect, in this experimental setup, f (lower stalk length is3,4,5mm; flowers1,2,3mm diameter) range, which when floret1mm diameter, the highest anther callus induction rate.2. The mountain of Rhodiola rosea floret1mm diameter, the most in the middle of mononuclear pollen development at this point, the anther callus induction rate in setting (below floret1mm diameter,1mm,2mm,3mm), within the scope of the highest then gradually decline.3. In three basic medium (MS, B5, N6) could anther callus induction rate was highest N6medium, however, in MS medium, anther callus growth rate faster, and the color is more green and yellow, and green dot.4. Auxin and cytokinin in culture there is a interaction relationship, when the concentration of auxin and cytokinin are low, the anther callus induction rate does not exist obvious differences between; When the concentration of auxin is changeless, cytokinin concentration rises, the anther callus induction rate does not exist obvious differences between; When the concentration of auxin and cytokinin are higher, the anther callus induction rate, the difference between this time, auxin using IBA, callus induction rate is higher. Auxin using NAA, callus piece of big, color is green. 5. In inoculation density10,20,30,40,50/bottle of inoculation density, the bottle could anther inoculation density, the greater the more conducive to anther callus induction.6. In this experiment set temperature (16℃,20℃,25℃and30℃), the temperature at25℃is conducive to alpine rhodiola anther callus induction; In the experimental setup of sunlight (10h,12h,14h,16h) within the scope of illumination time at12h could anther callus induction is more advantageous; The light intensity in this experimental setup (0lux,1000lux,2000lux,3000lux) within the scope of light intensity in the0-2000lux is conducive to alpine rhodiola anther callus induction.7. In the design of training environment, placed could anther inoculation after first under the condition of25℃dark train for7days, and then gives light, the callus grow faster in the growth process, and the color is mostly green, good overall situation of induction.8. In three basic culture medium (MS, B5, N6), the basic culture medium of MS, could anther callus with highest rate of green dot, B5middle and poor N6. In MS medium, anther callus differentiation of adventitious bud.9. The three kinds of auxin (NAA, IAA, IBA), auxin using NAA, alpine rhodiola anther callus green dot rate is higher, grow more adventitious bud, alpine rhodiola anther callus differentiation effect is good, and auxin IBA worst, auxin IAA for differentiation effect in the middle; In four kinds of auxin IAA concentration (0.1mg/L,0.5mg/L,1.0mg/L,2.0mg/L), are not conducive to high and low concentration could anther callus differentiation, in three kinds of cytokinin6-BA concentration (3.0mg/L,4.0mg/L,5.0mg/L), the callus differentiation rate increased with concentration present a rising trend; With four light time (10h,12h,14h,16h), illumination time of.12h, more conducive to alpine rhodiola anther callus differentiation.10. In the four kinds of light intensity (0lux,1000lux,2000lux,3000lux), light intensity for2000lux, more conducive to alpine rhodiola anther callus differentiation; In four kinds of temperature (16℃,20℃,25℃and30℃), when the temperature at25℃is for alpine Rhodiola anther callus.
