The major object of this study was to primarily establish the system of effective tissue culture of Osmanthus heterophyllus. The study took the fresh shoots and leaves as the explant,the optimal medium and culture programe were studied.The major results as following:(1) Using the fresh shoots of O. heterophyllus as the explant,the best season for selecting material was September and October.In this period,the live explant rate was hightest.(2) After being treated by alcohol for 90 seconds, the concentration of disinfectant mercuric chloride for stems was 0.1%. The optimizing time of disinfeetion was 5 minutes.The live explant rate was 77.5%.(3) After being treated by alcohol for 90 seconds,the concentration of disinfectant mercuric chloride for leaves was 0.1%. The optimizing time of disinfeetion was 9 minutes.The live explant rate was 87.3%.(4) The optimizing medium of initial culture was B5+1.0mg/LZT+0.05mg/LNAA+ 30g/Lsuorose.(5) Callus regeneration ability was different in different parts of the same leaf. The sequence of regeneration ability was:leaf base>leaf midst>leaf apex.(6) For leaves of Osmanthus heterophyllus inoculated on the far axial surface,the induction rate was above 90%,but for the ones inoculated on the paraxial surface, their callus tissue could not be organized.(7)The optimizing medium for callus induction was B5+0.5mg/L 6-BA+1.0mg/L 2,4-D + 30g/Lsuorose.(8)The effects of light and dark on callus induction had no significant different.The quality of callus formed in dark was better than light.(9)The optimizing medium was B5+3.0mg/LZT+30g/Lsuorose in the multiplication of buds. The average propagation coefficient was 3.63.(10)The optimizing medium of rooting was B5+3.0mg/LNAA+ 30g/Lsuorose.The rooting rate was 63.3% and the average numbe of root was 3.7.(11)The optimizing soil as culture substrat was Pearlite:Vermiculite:Peat=1:2:1,the ratio of transplant viability was above 93.3%.
