This research takes wild Pinellia ternata as material, by adopting quadrature floc test, using the leads of induction, enzyme amplification and the seedling rate for index mark, we take a comprehensive critique to the tissue culture of Pinellia ternata, and do research on the influence to the endogenous hormones, the solubility protein contents, peroxide Mao(POD) and benzene, alanine crack Mao(PAL) and change of albumen Mao's activity by adding endogenous hormones hormone in the course of tissue culture of Pinellia ternata. And also do an inquiry into the correlation of cell divides, organ occurrence, physiology bio-chemical change with development condition, in the process of the one step becomeing seedling of Pinellia ternata. The main conclusions as follows:1,The tuber is the superior outsider plant corpus in the process of quasing one step becomeing seedling of Pinellia ternata by adopting quadrature floc test.2,By synthesizing the leads of induction, enzyme amplification and the seedling rate three index marks, the best hormone proportioning combines of the one step becomeing seedling of Pinellia ternate is MS+0.2mg/L2,4-D+1.0mg/LKT+0.1 mg/LNAA.3,The live rates of transferase of tissue culture seedling of Pinellia ternata are all higher. The tuber tissue culture seedling in the best hormone proportioning culture medium and those tuber tissue culture seedling that are added nitrogen(niter 5.5g, 6.0g,6.5g), there live rates lead to 100%. The live rates of transferase of blade and petiole tissue culture seedling in the best hormone proportioning culture medium are all above 90%. The transferase plants into the quasi Pinellia ternata seedling for living will also utter new seedling.4,Detecting through a quadrature floc test, we find the inside source IAA number of the tissue culture of Pinellia ternata those are on the culture medium res MS+0.2mg/L2,4-D+1.0mg/LKT+0.1 mg/LNAA hit high peak on the 14th day or the 21th day. The other's inside source IAA numbers are aways latter than this handing, it is on the 28th day or the 35th day. The height of the high peak will make influence on uncertain bud divides, this explains that the pangene formings need the accumulatting of somatotropin to start.5,Through the research on the relationship between inside hormone in the tissue culture res of Pinellia ternata on different endogenous hormones hormone proportioning culture medium and morphogenesis, it is explained that:in various of handling, the occurrence of treating wound structure of uncertain bud have something to do with the height of inside source IAA contents. When the inside source IAA come to the lowest, none uncertain bud generate. The creation and formulating of uncertain bud need to undergone a step down course of inside source IAA. The course of occurrence of treating wound structure of uncertain bud always accompany with the droop of inside source CTK contents.6,In the process of developing, the research on the solubility protein contents, peroxide Mao(POD) and benzene alanine crack Mao(PAL) and change of albumen Mao's activity in the tissue culture res of Pinellia ternata, it is explainted that:The protein level, the albumen Mao activity variance has something to do with the growth of treating wound organ, but it can't be the sign of cell divides. The formulating of organ has close relationship to POD and PAL activity. The live law of POD in treating wound organ are different in the Morphogenesis course. The changing law of PAL activity are consistent in the morphogenesis of treating wound organ,and its activity droops along with the formulating of uncertain bud, and also play an important role in the indogen divides.7,The total alkaloid contents in the tuber of Pinellia ternata which become to live after chaining the tissue culture seedling of outsider plant corpus of Pinellia ternata are higher than those in the tuber of wild Pinellia ternata.After adding niter to the culture medium, it can not only raise the total alkaloid contents of Pinellia ternate, but cut low its contents. The total alkaloid contents in the tuber of Pinellia ternata which have been added 5.5g niter to the culture medium are higher than those added nothing.8,The looks of the HPLC diagram are consistent through this test.By using various data handling methods,the result of the analyse marks that the tubers of the tissue culture seedling on the outsider plant corpus, the tuber of the tissue culture seedling on the outsider plant corpus and the tuber of wild Pinellia ternata have some reciprocity. Tissue culture Pinellia ternata that have been added nitrogen also have some reciprocity with those have not been added. Its chimical composition is more stable and consistent.
