As a traditional Chinese medicine resource with a history of more than 2000 years,Pinellia has obvious antitussive and expectorant effects,promotes bile secretion and enhances intestinal transport capacity.Development has a broad market and prospects.This paper focuses on the difficulties of Pinellia ternate bud reproduction,we first carried out bud grade grading and response surface methodology to optimize germination conditions,starting from the physiological and biochemical changes of storage substances,amino acids,endogenous hormones and phenols during germination The research explored the suitable germination mode of Pinellia ternata buds and the corresponding physiological and biochemical changes,providing scientific basis and technical support for solving the problem of Pinellia ternata buds breeding,and mainly achieved the following main conclusions:(1)According to the diameter of bead buds,three grades are set,among which grade I bead is more than 0.6 cm,grade II bead is 0.3-0.6 cm,and grade III bead is 0.3 cm or less.The germination rate of Pinellia ternate buds is positively correlated with the size of the bead buds.The larger the bead buds,the higher the germination rate.The germination rates of Pinellia ternate III,II,and I are 80 ~ 92%,53 ~ 61%,23 ~ 31%.(2)Aiming at the problem of Pinellia ternate bud propagation difficulty: Pinellia ternate bud germination difficulty is mainly due to the difficulty of small bead bud germination.Under the artificial culture conditions of this experiment,about 12 days can develop into normal seedlings smoothly.Through the design-Expert.8.05 b software to solve the regression equation of germination rate,the best conditions for the germination of Pinellia ternate buds are: A:germination temperature 22.6 ℃,B: soil moisture 17.8%,C: 6-BA concentration 15 mg / L,D:NAA concentration 0.51 mg / L.Through the production method of artificial bud germination of Pinellia ternate,the ability of Pinellia ternate bud breeding can be enhanced,which helps to promote Pinellia medicinal materials to completely get out of the resource predicament.(3)Division of bud germination stage: According to the morphological changes of the bud germination process,it is mainly divided into five stages: dried bead bud,imbibed bead bud,dew bead bud,elongated bead bud,and green bead bud.According to the overall difference in metabolic profile,it can be divided into 3 stages The main stages are: dry bud stage,bud germination stage,and seedling establishment stage.(4)Physiological law of bead bud germination process: Pinellia ternate bud germination process involves a wide range of metabolic regulation behavior.In this experiment,24 metabolite components detected by HPLC and UV showed significant content changes during the germination process.Starch,ABA and other contents showed a downward trend,ZT,IAA,soluble sugar and other contents showed an upward trend and then a downward trend,may involve 10 key metabolic pathways,respectively: glycine,serine and threonine metabolic pathway,butyrate metabolism Pathway,alanine,aspartate and glutamate metabolism pathway,aminoacyl-tRNA biosynthesis pathway,arginine and proline metabolism pathway,phenylalanine metabolism pathway,tryptophan metabolism pathway,flavonoids And flavonol synthesis pathway,valine,leucine and isoleucine biosynthesis pathway,flavonoid biosynthesis pathway.
