Fatty acid binding proteins (FABPs) are made up by A-FABP,H-FABP,B-FABP,M-FABP,E-FABP,IL-FABP,L-FABP,T-FABP and I-FABP.FABPs are considered to be important carriers for the transportation of intracellular fatty acid. They transport fatty acids from the plasma membrane to sites of fatty acid oxidation, of fatty acid esterification into triacylglycerols or phospholipids, or to the nucleus for possible regulatory functions. FABPs play an important role in the modulation of lipids metabolism. In the present study, many scholars have studied about polymorphysims of A-FABP gene, but it's only a few locuses existed in exon and intron areas. Our study also detected many variable sites in the 5'region and 3'region besides the exon areas. The 10th generation populations from NEAU (Northeast Agricultural University) broiler lines divergently selected for abdominal fat content (NEAUHLF), NEAU-F2 and AA broiler random population were used. Sequencing, PCR-RFLP,PCR-LP,DHPLC methods were developed to detect polymorphisms of candidate genes; the htSNPs were chosen to construct haplotypes; and then the individual SNPs and haplotypes of A-FABP associated withchicken growth and body composition traits. The main results were as follows:(1)Based on DNA sequencing, PCR-RFLP,PCR-LP,DHPLC polymorphisms of 5'region of A-FABP,5 SNPs were detected by sequencing for the 10th population of the NEAUHLF. (g.-1376 G>A,g.-1117 C>T,g.-972_-963 del 10, g.-846_-845 ins 22,g.-415_-409 del 7), three SNP were detected in exon 1,exon 3 and 3' region(g.51 C>T,g.1729G>A,g.3163T>C), respectively. Linkage disequilibrium (LD) analysis revealed g.-1376 G>A,g.-1117 C>T,g.-972_-963 del 10,and g.-846_-845 ins 22 polymorphisms sites of A-FABP gene exist in a steady haplotype block, g.-415_-409 del 7, g.51 C>T, g.1729G>A exist in another steady haplotype block. g.-1376 G>A, g.-1117 C>T,g.-972_-963 del 10,g.-415_-409 del 7,g.3163 T>C were chosed as htSNPs. LD analysis revealed g.-972_-963 del 10,g.-846_-845 ins 22,g.51 C>T,g.1729 G>A polymorphisms sites of A-FABP gene exist in a steady haplotype block in NEAU-F2 population; and g.-846_-845 ins 22,g.1729 G>A were chosed as htSNPs.(2) Based on DNA sequencing, PCR-RFLP,PCR-LP,DHPLC polymorphisms of the 8 SNPs of A-FABP gene, the 10th generation population of the Northeast Agricultural University broiler lines divergently selected for abdominal fat content was used. The different genotype frequency and allele frequency were analysised in 10th generation of the NEAUHLF; and chi-square test was detected for allele frequency. The results showed that whatever in the fat line or the lean line, the genotypes distribution were not balance, specifically for the g.-415_-409 del 7 locus, theâ…¡genotype was really rare, only 9 birds withâ…¡genotype. The results of allele frequency showed that the 6 SNPs except for g.-1117 C>T,g.-972_-963 del 10 had highly significant difference (P<0.01).(3)Combination the results of the eight individual SNPs and haplotypes analysis showed that there were only significant relation with muscle stomach weight (MSW) and percentage of muscle stomach weight (MSW/BW) (P<0.05) in The 10th generation population of the Northeast Agricultural University broiler lines. The results of the four individual SNPs and haplotypes analysis showed that there were significant relation with body weight at 6 wk (BW 6),metatarsus length at 12 wk (MeL),spleen weight (SW),muscle stomach weight (MSW),percentage of muscle stomach weight (MSW/BW) and metatarsus claw weight (MeCW) (P<0.05),and highly significantly correlated with keel length (KeL),femur length (FeL),pectoralis minor weight (PMiW) and percentage of pectoralis minor weight (PMiW/BW) (P<0.01)in the NEAU-F2 population. The results of the two individual SNPs showed that there was no significant relation with all the traits at g.-972_-963 del 10 locus (P>0.05);there were significant relation with body weight at 5 wk (BW5) and spleen weight (SW) (P<0.05) at g.-846_-845 ins 22 in AA broiler random population.
