Objective: It was studied the protective effect of phenolic alkaloids from Menispermum dauricum(PAMD) on experimental cerebral ischemia in rats and explored the mechanism of action. Methods: The imcomplete cerebral ischemia was induced by the occlusion of bilateral common carotid arteries(CCA), 45min after ischemia ,the occlusion was removed and the blood was rapidlly reinfused. Water content, superoxide dismutase (SOD) activity, malondialdehyde(MDA) and nitric oxide(NO) content of brain tissue were determined and observed the effect of PAMD on thrombosis formation;The focal cerebral ischemia model was produced by occluding middle cerebral artery(MCA) for 2h and reperfusing 24h.Cell apoptosis was detected by Tunel,the expression of bcl-2 and bax were analyzed by immunohistochemical.ResuIts:PAMD can decrease water content of brain tissue,reduce producion of MDA and NO,enhance activity of SOD and inhibit thrombosis formation;reduce cell apoptosis after ischemia reperfusion,induce bcl-2 expression and inhibit bax expression.Conclusion:PAMD processes protective effect on experimental cerebral ischemia.The mechanism of action is relate to reduce production of free radical and NO,anti-thrombosis formation and inhibit cell apoptosis.
