Backgrouund:Pathological scars ,including hypertrophic scar and keloid ,are the result of over wound-healing,characterized by aberrant proliferation of fibroblasts and the accumulation of excessive collagen and extracellular matrix,affected the function and cosmic reconstruction of a patient.The pathological mechanisms underlying the two lesions is not clear,recent studies have suggested that genes dysregulation is a key event.E2F family of transcription factors play a key role in G1-S phase ,cell cycle regulation and cell proliferation. Previous studies show that overexpression of E2F1 promote cell prolifertion, it is involved in the process of carcinogenesis. Pathological scar has similar molecular mechanisms with tumor, the relationship between pathological scar and E2F1 gene has not been reported. Objective:In order to inverstigate the correlation between the expression of E2F1 gene and role in tumorigenic process and abnormal scars compared with normal skin and normal scar,and to elucidate the role of E2F1 in tumorigenic process. Methods:1.The E2F1mRNA of tissues was investigated in the specimens from normal skin, normal scar, hypertrophic scar and keloid tissues using reverse transcription polymerase chain reaction(RT-PCR). 2. Normal skin, normal scar, hypertrophic scar and keloid tissues fibroblasts were cultured, the distribution of cell cycle were analysised with flow cytometry, the E2F1mRNA of fibroblasts was detected by RT-PCR. 3.Immunohistochemiscla technique was performered to detectthe expression and distribution of E2F1 protein in the paraffin embedded tissues samples from patients,statistics was used to analyze the date.Result:1.The expression levels of E2F1mRNA in hypertrophic scarand keloid was much higher than normal skin and normal scar; 2.The propagative fibroblasts derived from hypertrophic scar and keloid was significantly elevated in comparison to the normal skin and normal scar , E2F1mRNA expression in fibroblasts was also higher than control groups; 3.The positive rate of E2F1 protein expression was no distinct difference between the hypertrophic scar and keloid ,there was remarkably significant compared with normal skin and normal scar(p<0.01). Conclusion:The result indicate that the expression of E2F1 was increased in the hypertrophic scar and keloid, E2F1 overexpression may play an important role in the proleferation of fibroblasts and in the development of pathological scar.
