Chapter I Distribution and expression of bone morphogenetic protein -2/4 in keloidObjective: To explore the role of bone morphogenetic protein in the pathogenesis of keloid.Methods: Specimens of 6 normal skin, 6 hypertrophic scars and 6 keloid lesions were harvested and the distribution and the expression of the BMP-2/4 was examined by immunohistochemistry.Results: Immunohistochemical results demonstrated that the MG of BMP-2/4 in keloid lesions and hypertrophic scars were significantly smaller than that in normal skins(P<0.01)and no obvious difference in the two types of scars(P>0.05).Conclusion: 1. Positive staining of BMP in keloid, hypertrophic scar and normal skin was mainly in the epidermal cells, dermal matrix, hair follicle and flbroblasts. 2. The expression of BMP-2/4 in different levels during the process of wound healing might be related to the formation of keloid lesions.Chapter II The expression of BMP-2 in rabbit skin of deep second degree scald woundObjective: to study the change of endogenous bone morphogenetic protein in rabbit skin of scald wound.Methods: Either side of the back midline was scalded to deep second degree and the other side of normal skin as control in 30 adult rabbits. Every 6 rabbits were killed in 6, 72h, 1, 2 and 4weeks respectively after operation and skin specimens were immunohistochemically examined for BMP-2 expression.Result: in the control skin the gray level of BMP-2 was as much as the gray level of BMP-2 after operation(P>0.05).Conclusion: BMP-2 may not play a role in the early irritable actions in scald wound.
