IntroductionThe recent research show: nasal polyps are the results of nasal mucosa chronic inflammation, which related to multiple factors, inflammatory medium play an important role in the development of nasal polyps. Cyclooxygenase(cox) is an crucial enzyme in the metabolism of arachidonic acid to prostaglandins(PG) , thromboxanes and prosta-cyclin. It is now clear that COX - 1 is expressed in the majority of cells constitutively, and COX -2 appears after stimulation. ,otherwise Cyclooxygenase - 2 can regulate the expression of many inflammatory medium . This artrcle is going to explore the relationship between COX -2 and nasal polyps.Objective : explore the relationship between COX - 2 and the oc-curance, development and classification of nasal polyps by testing the expression of COX - 2 in nasal polops and normal nosepiece.Materials1: resource of specimens: specimens from 26 cases of nasal polyps which had been diagnosed by pathology havenot be treated by steroid before operation, and all the patients without the histoy of hyper-susceptibility. The specimens are divided into 2 groups basing on the results of nasal cavity examine and CT. Group A includes 14 case ofI model and II model 1 ^2 expect nasal polyps. Group B includes 12 cases of H model 3 expect and HI model nasal polyps. Ten of the specimens comes from the nomal nosepiece and all the patients without the histoy of hypersusceptibility and chronic rhinitis.2: major agents and equipments; ariti - COX - 2 ( purchased form Wuhan Beshide company). Micromage .analysis system.MethodsThe specimens from 26 nasal polyps case and 10 normal nose-piece are fixuped for 48 hour in 10% formalin and then embed in ole-fm,made into 4u,m olefin slice up. and dye them with immuneohisto-chemical method. The slice up are observed with photicsmicroscope, the positive results standared by cytoplast and karyotheca appearing orange. The results are analysied with Computer microscope image a-nalysis system get the ratio of positive areas, put the data into Spss software to analyse .ResultsCOX - 2 positive rinse can hardly be found in the group of normal nosepiece and negative contrast group , but it can be observed in the group of nasal polyps . positive COX - 2 in nasal polyps located in lymphocyte, serouscyte and epithelium. Analysis with spss soft ware find the expression of COX - 2 in nasal polyp tissue obviously highen than in nomal nosepiece(P <0.01) . and COX -2 expression in Group B is higher than it in Group A( P <0.05).DiscussionRecent research show; inflammatory medium play an important role in the development of nasal polyps. COX - 2 is one of the important inflammatory medium. It is not only an crucial enzyme in the metabolism of arachidonic acid to prostaglandins(PG) , but also regulate the expression of many inflammatory medium, it closely related to the occuronice, development of nasal polyps . the result of the experiment show, the positive area ratio in nasal polyps obviously higher than it in normal nosepiece( P < 0.01). and the COX - 2 positive area ratio in Group B is higher than it in Group A ( P < 0. 05 ). all these suggest that COX - 2 play an important role in the development of nasal polyps by regulating the compose and activity of prostaglandins and other inflammatory medium .ConclusionHigh expression of COX - 2 in nasal polyps may play an important role in the pathogenes of nasal polyps.
