Objective Intercellular adhesion molecule-1 (ICAM-1, CD54) and vascular cell adhesion molecule-1 (VCAM-1, CD 106) are members belong to immunoglobulin supeurfamily (IGSF) of cell adhesion molecules. Their ligands are lymphocyte function antigen-1 (LFA-1, macrophage antigen complex-1 (MAC-1) and very late antigen-4 (VLA-4) respectively. The members of IGSF not only have great significance in modulating immune function of bodies but also participate in attachment and recognition of cells. In recent years, ICAM-1 and VCAM-1 which are found in serum are called soluble ICAM-1 and VCAM-1 (sICAM-1, sVCAM-1), they include all the extracellular domains and have capability to joint with their corresponding ligands. sICAM-1 and sVCAM-1 can affect immune state of bodies and participate in immune " escaping" actions of tumor cells by inhibiting biological actions of ICAM-1 and VCAM-1. They inhibit adhesive functions by preventing the ligands on membrane of tumor cells from binding to their corresponding receptors. This study is to observe the significance of sICAM-1 and sVCAM-1 in diagnosis, the development of clinical status, treatment outcome and prognosis of acute leukemia patients by investigating their serum levels.Methods Serum samples were obtained from 19 acute lymphocytic leukemia (ALL) and 36 acute non-lymphocytic leukemia (ANLL) patients before and after treatment, 6 relapsed ALL and 10 relapsed ANLL patients. All serum samples were measured for concentrations of sICAM-1 and sVCAM-1 with enzyme linked immunosorbent assay (ELISA) method.Results Serum levels of sICAM-1 and sVCAM-1 in acute leukemia patients elevated significantly before treatment compared with normal control group and reduced to normal levels after remission. Serum levels of sICAM-1 and sVCAM-1 in relapsed acute leukemia patients also elevated significantly compared with normal control group. Serum levels of sVCAM-1 in M4 and MS are higher than other subtypes in ANLL patients. Patients with elevated serum levels of sICAM-1 and sVCAM-1 hadpoorer treatment outcomes. Serum levels of sICAM-1 and sVCAM-1 were found to return to normal at remission through ambulatory observation.Conclusion It has a certain help to measure serum levels of sICAM-1 and sVCAM-1 in acute leukemia patients for diagnosis and judging the development of clinical status, treatment outcome and prognosis.
