Objective: To investigate the expression of VEGF, CD44v6 and Ki67 in non-small cell lung carcinoma and their correlation with growth, metastasis, cellular prolific activity and lymph node metastasis. Methods: VEGF,CD44v6 and Ki67 expressions in 68 cases lung cancer tissue (62 cases NSCLC, 6 SCLC) and 20 matched normal lung tissues were studied by immunohistochemical Staining. Survival curves were platted using the method of Kaplan -Meier, and the log-rank test wasused to determine statistical differences between life tables. A Cox proportional hazard model was used to assess the effect of patient and tumor variables on overall survival and of tumor variables on nodal involvement.Results: 1. The positive rates of VEGF, CD44v6 and Ki67 in NSCLC were 72.6%, 69.4% and 80.6%, respectively. These all were higher than matched normal lung tissues. P<0.05. 2. Their expressions were not related to years, sex of patients, and the histological classification in NSCLC. 3. The expression of the three markers was related to clinical stages and the metastasis of lymph nodes. The expression rates of the three markers in NSCLC with the metastasis of lymph nodes were significantly higher than without metastasis, p<0.05, and with Ⅲ-Ⅳ stages were significantly higher thanⅠ-Ⅱ Stages, P<0.05. 4. The expression of VEGF was related to the expression of Ki67(χ2 =26.6), No associations were found among the CD44v6 and VEGF, Ki67(χ2 =0.43 and 0.475).5. Survival rate of three years were significantly higher to those patients with the positive expression of these three makers than those with the negative expression, P<0.05. 6. In a multivariate analysis, TNM staging and the metastasis of lymphnodes were the independent prognostic factors, VEGF, CD44v6 and Ki67 were not an independent factor that impact on the prognosis.7. Kaplan-Meier survival curves showed a statistically significant worse prognosis for patients with high VEGF, CD44v6 and Ki67. Conclusion: The expression of VEGF, CD44v6 and Ki67 have a significant relation with microssel formation, the growth, invasion of carcinoma and lymph node metastasis. But CD44v6 and VEGF, Ki67 do not pass same course to these result. So, the formation and metastasis of NSCLC are a complicated course. These patients of NSCLC with higher expressions of VEGF, CD44v6 and Ki67 may have poor prognosis.
