Objective To investigate the expressions of human heparanase (HPA) and non-metastasis gene (nm-23) in Non-small cell lung cancer, and explore the relationship between their expressions and tumor’s clinicopathological characteristics. Methods The expression of HPA mRNA and nm23-H1mRNA in NSCLC cancer tissues(n=48), tumor adjacent tissues(n=10) and normal lung tissues(n=10) were detected by realtime reverse transcription polymerase chain reaction(realtime RT—PCR). Results The expression level of HPA mRNA(8.244±5.314) was markedly higher than that in tumor adjacent tissues(1.043±0.478) and normal lung tissues(1.025±0.465)(P<0.01). The high expression level of HPA mRNA in the NSCLC tissues had significantly correlated with the pathology types (P<0.05), lymphatic metastasis (P<0.05) and TNM stage (P<0.01). No significant differences in high HPA mRNA expression were observed with smoking, age, gender and degree of differentiation among the NSCLC patients (P>0.05). The mean expression level of nm23-H1mRNA(0.599±0.566) was markedly lower than that in tumor adjacent tissues (1.232±0.328) and normal lung tissues(1.443±0.427)(P<0.01).The low expression level of nm23-Hl mRNA in the NSCLC tissues correlated significantly with the pathology types (P<0.05) lymphatic metastasis (P<0.05),TNM stage (P<0.01) and degree of differentiation. No significant differences in low nm23-Hl mRNA expression were observed with age, smoking, gender among the NSCLC patients (P>0.05). The high expression of HPA mRNA had positive correlation with the low expression of nm23-Hl mRNA; Conclusion HPA and nm23-Hl gene are closely related to oncogenesis, development, and metastasis of human NSCLC, the two gene may be two important biological markers in reflecting the progression, infiltration, metastatic potential,clinical prediction and prognosis of NSCLC and provide theoretical basis of gene targeted therapy.
