Gengeng-Qinlian Decoction(GQD) is a famous prescription by extraordinarily skilled practitioner Zhangzhongjing, which is composed of Pueraria lobata Ohwi, Coptis chinensis Franch, Scutellaria baicalensis Georgi and Glycyrrhiza uralensis Fisch. It can relieve exterior syndrome, remove obstruction due to interior syndrome, ascend the clear, stop diarrhea and has been widely used in clinical routine. But to GQD, the effective assay methods is lack and the study on compatibility is deficiency too.Objective: To develop the assay methods of the effective components(EC) and the fingerprint (FP) of GQD. To study the influence of compatibility on the content of EC and FP.Method: After a simple sample extraction with methanol solution, high-performance liquid chromarography(HPLC) was adopted to assay the content of EC and high -performance capillary electrophoresis(HPCE) was adopted to assay FP. Orthogonal design was used in this research and the data was analyzed with statistics analysis(SPSS 10.0).Result: (1) .All these methods were simple, accurate and reproducible. (2) For Scutellaria baicalensis Georgi and Glycyrrhiza uralensis Fisch, the difference on the content of kakonein, and the difference on the peak area of the Pueraria lobata Ohwi's G1, G2, G3, G5, G6 peak in FP were insignificant(P>0.05);(3) For Pueraria lobata Ohwi and Scutellaria baicalensis Georgi , they may reduce the content of berberine and the peak area of the Coptis chinensis Franch's L5 peak in FP(P<0.05); the difference on the peak area of the Coptis chinensis Franch's L5 peak in FP was significant(P<0.05) , but the difference on the peak area of the Coptis chinensis Franch's L1 and L4 peak in FP was insignificant(P>0.05). For Glycyrrhiza uralensis Fisch , it may reduce the content of berberine, and the difference on the peak area of Coptis chinensis Franch's L1,L4 and L5 peak in FP were insignificant(P>0.05).(4) For Pueraria lobata Ohwi, it may reduce the content of baicalin(P<0.05), and the difference on the peak area of Scutellaria baicalensis Georgi's Q1, Q2, Q3 and Q4 peak in FP were insignificant(P>0.05).For Coptis chinensis Franch,it may reduce the content of baicalin and the peak area of the Scutellariabaicalensis Georgi's Q4 peak in FP(P<0.05). For Glycyrrhiza uralensis Fisch,the difference on the peak area of Scutellaria baicalensis Georgi's Q1, Q2, Q3and Q4 peak in FP were insignificant(P>0.05).(5)For Coptis chinensis Franch, it may reduce the content of glycyrrhiza . ForScutellaria baicalensis Georgi and Pueraria lobata Ohwi, the difference onthe content of glycyrrhiza was insignificant(P>0.05).(6) When Coptis chinensis Franch decocted with Scutellaria baicalensisGeorgi, Pueraria lobata Ohwi or(and) Glycyrrhiza uralensis Fisch, thesedimentation appeared, berberine, baicalin, kakonein and glycyrrhiza werefound in it, it was proved that the main component were berberine andbaicalin.Conclusion: The the content of EC and the FP of GQD.were influenced bycompatibility. Sedinentation appeared when Coptis chinensis Franch wasincluded in the compatibility, the main reason is the change of pH leads tosedimentation in decoction and reducing of some components.
