Objective: The last few years, much evidence suggests that free radicals damaging mechanisms are important in acute ischemic stroke. Free radicals attack important macromolecules leading to cell membrane and alter their fluidity characteristics, and also lead to release of potentially toxic byproducts. Free radicals react with proteins leading to enzyme inactivation mutation, disruption of transcription and translation. To clear the free radicals and to breakdown their chain reactions may play an important therapeutic role in cerebral ischemic injury. PD is one of the active components isolated from Polyonum Casplatum Sieb.et zull. This study is to explore the scavenging effect on free radicals of PD in the experiment study of focal cerebral ischemic and to provide experimental evidence for futher research and tap of cerebral protective medicines.Methods: The animal model of the right MCA ischemia was established by suture method in mature wistar rats. Twenty four mature male wistar rats. weighing 200-250g were chosen. The animals divided into three groups randomly: Sham-operated group, ischemia model group. PD treatment group. We observed the changes of malonaldehyde(MDA), superxides dismutase (SOD),Nitric oxide synthase (NOS),glutathione peroxidase (GSH-Px),catalase(CAT),water content in brain and pathological changes after cerebral ischemic 2h.Results: 1. The content of MDA and NOS activity increase and SOD, GSH-Px,CAT activity decrease in the ischemic model group, which has significance compared with sham operated group(P<0.01).2. The content of water increase in the ischemic model group, which has significance compared with sham operated group(P<0.01). HE of ischemic brain tissue demonstrate the brain edema. 3. PD can reduce MDA level and increase SOD, GSH-Px, CAT activity in the ischemic brain tissue, which has significance compared with ischemic model group(P<0.01).4. PD can reduce water contents in the ischemic brain tissue, which has significance compared with ischemic model group(P<0.01). HE demonstrate that PD can reduce the brain edema.5. PD can reduce NOS activity in the ischemic brain tissue, which has not significance compared with ischemic model group(P>0.05).Conclusion: PD could lower the level of MDA, improve the activity of SOD, GSH-Px, CAT, reduce brain edema. Our results that prophylactic use of PD has protective effect on focal cerebral ischemia in rats, and its treatment mechanism maybe come from its effectively scavenging free radicals.
