Objective: We have studied the effect of the different frequency of electron-acupuncture on the rate's ciatic nerve regeneration after transection injuried .By observing the morphological and functional changes during the regeneration, we want to select the optimum parameter and the best therapy time.Method: 150 rats were divided into control group, model group, 5Hz group and 100 Hz group at random. Except the control group ,the sciatic nerves of other groups were transectioned and then bridged the two stumps into the silicon tube to form a regeneration chamber. There was a 5mm defect between the two stumps. 5 days later , to acupuncture the Huan Tiao, Zu Sanli and San Yinjiao acupoint at the affected side limb for 10 minutes every two days with different frequency. Postoperative survival times were 14, 17 and 20 weeks. The nervous tissue were dealed with HE staining, Sevier-Munger staining, Page staining and immunohisto demistry , the superstructure of regeneration tissue were observed by electron microscope at the 20th weeks. The muscle filter diameter and section area of gastrocemius were analysised by IPP system. We also studied the EMG of gastrocemius which reflected the function recovery of injured sciatic nerves.Results:1 .The nerve regeneration of 5Hz groups is the best at all different therapy time points. The S-100 protein expression in 5Hz group is higher than that of 100 Hz and model groups ,( P<0.05). It is also higher in 100 Hz group than that of model group at 14 weeks. But there was no statistics significant between l00Hz group and model group at 17 weeks and 20 weeks. The result of the electron microscope shows that 5Hz electron acupuncture group can improve the recovery of nerve cell membrance structure and other superstructure.We can say that 5Hz electron acupuncture can promote nerve regeneration. 2.The recovery of muscle fiber diameter and transection area in 5Hz group is better than in and model group. At 14,17 and 20 weeks after sciatic nerve injuried (P<0.01). It is also better in lOOHz group than that in model group at14 and 17 weeks(P<0.05).The results illustrate that the EA can remarkably defer the gastrocemius asttophy.3.At different therapy time points ,there is a tendency of recanclization rate that the 5Hz groups is higher than l00Hz groups than that model groups. But there is no statistics significance.Conclusion: The EA therapy can effectively promote the rat' sciatic nerve regeneration,especially the low frequency effect is the best, which need advanced study.
