Objective: To select the optimum therapy time and parameter on regeneration of the injured peripheral nerve treated with electro acupuncture (EA) at different frequencies or low frequencies in different time.Method: The sciatic nerve was crashed in the Wistar rat to construct Nerve Regeneration Chamber, and all of the rats were randomly divided into healthy control group, model group, 5 Hz-EA group, changed frequency group I and chenged frequency group II. EA was applied 3 times per week to Huantiao, Zusanli and Sanyinjiao for 10 minute 5 days after crush. Nerve regeneration was observed 13 and 20 weeks later with HRP tracing, HE staining, Sevier-Munger staining, S-100 immunohistochemical test, solochrome cyanine R staining and electron microscope scanning. The recovery of sural muscle was detected by H E staining and sectional area measuring of muscle cell with photo analysis system.Results:1. The amount of HRP labeled cells were most in control group, and the amount EA treated groups were between control and model group in 13 and 20 week.2. In EA treated groups, the expression of S-100 increased significantly in 13 week (P < 0.05, P<0.01), while the effect of 5 Hz-EA group and changed frequency group I were better than changed frequency group II. Such is the case in 20 week, but there was no significant difference among different frequency group II, control group and model group. The data showed that there was no significant difference in each group in two time point. The SC hyperplasia, myelin sheath arrangement, nerve fiber growth and ultrastructure in EA treated groups were better than model group but didn't be the same with control.3. In 13 week, the recovery of sural muscle was better than model group (P < 0.01), while there was no significant difference among EA groups. But in 20 week there showed different things, the effect in 5 Hz-EA group and changed frequency group I were better than model but no difference between themselves. In contrast, changed frequency group II had the same effect with model. In addition, there do the same effect in each group in two time point, but had significant difference to control.Conclusion: The EA treatment can promote the regeneration of the rat'speripheral nerve, espscially, 5 Hz and changed frequency I EA treatments do better changed frequency II EA.
