| Objective:To discover possible the mechanisms of restraining the growth of tumor and its anti-angiogenesis of Xiaotan Sanjie Decoction by way of observing the expression of angiostatin and endostatin on human gastric cancer tissue on MKN-45 nude mouse model。Methods:experimental MKN-45 nude mice had been randomly divided into saline group,5-Fu group, Xiaotan Sanjie Decoction group randomly after the orthotropic implanted model bearing gastric carcinoma cell line MKN-45 The administrations began from the 48 hours after the surgery, which were as follows:the mice in saline group were given isotonic saline 0.4ml ig. once a day×6w; in 5-Fu group were given 5-Fu dilution(60mg.kg-1.w-1) 0.2ml ip. once a week×3w; in Xiaotan Sanjie Decoction group were given Xiaotan Sanjie Decoction 0.4ml ig. once a day×6w, The experiment terminated on the 6th week. The mice were sacrificed after abdominal cavity anesthesia, observe the gastric partial growth of transplanted tumor, tumor tissue taken, Immunohistochemical staining of CD34, counted MVD, quantitative observation of tumor angiogenesis, while detected angiostatin and endostatin protein and mRNA of tumor tissues by immunohistochemistry and RT-PCR Results:Xiaotan Sanjie Decoction can inhibit the growth of orthotropic implanted tumor, reduce its blood supply, and a significant reduction in tumor tissue MVD, significantly increases the expression of angiostatin and endostatin protein and mRNA. Conclusion:It might be one of the mechanisms of Xiaotan Sanjie Decoction that can increase the expression of angiostatin and endostatin proteins and mRNA in gastric tissue by restraining the growth of tumor and anti-angiogenesis.
