Objective To investigate the changes of tumor cells kinetics after radio frequency ablation and to provide an experimental basis for multidisciplinary therapy.Methods Balb-C mice were subcutaneously implanted with H22 cells to establish ectopic models of liver cancer. The models were treated by radio frequency ablation. The changes of tumor cells kinetics were analyzed by flow cytometry. Proliferation and apoptosis of tumor cells were examined by immunohistochemistry.Results 1. Tumor nodus were established subcutaneously in Balb-C mice and no tumor metastasis was detected. The tumor size was about 2.0 cm and the mice health status was acceptable 23 days after inoculation, which was considered a suitable time to undertake radio frequency ablation. 2. The radio frequency ablation condition was setted as 10W, 60℃ and 10min to ensure central coagulability necrosis and sufficient residual tumor tissue in the mean time. After radio frequency ablation, the cell cycle of the local part and separate part changed with time. S stage cell proportion increased and the prohferation index(PI) enlarged. 3. Proliferation and apoptosis of tumor cells were gradually enhanced together at both local part and separate part. There was significant difference in the apoptosis index(AI) among different parts(P<0.05). AI of the local part was higher than the separate part, and AI was the highest in the matched region of local part. There was significant difference in the PI among different parts(P<0.05), PI of the peripheral region of local part was the highest.Conclusion Tumor cells kinetics could be changed by radio frequency ablation. The enhancement of proliferation appeared at both local part and separate part. Effect of nerval system and endocrine system could not be neglected. The increment of S stage cell proportion indicated radio frequency ablation could provide convenience to the next therapy, and afte radio frequency ablation the curative effect of generalize...
