Coronary Heart Disease (CHD) is a kind of multifactor diseases that include environmental and inherent factors . Several kinds of dangerous factors appear simultaneously and dynamically interact on each other. Recently, gene polymorphism has been a new field in medical research. Because of varies in dangerous allele, the tendency of thrombosis is extremely different among races and areas. Objective To examine the matrix metalloproteinase-9 (MMP-9) C-1562T and platelet glycoprotein IIIa (GPIIIa) T1565C mutation in Shanxi province and study the relationships of these mutation with CHD. The level of plasma fibrinogen(Fg), total cholesterol (TC), triglyceride(TG), low density lipoprotein(LDL), high density lipoprotein (HDL), lipoprotein a (LPa), apoliprotein A(APOA) and apoliprotein B(APOB) have been measured and to explore their clinical significance. In the study, logistic regression has been used to analyze the dangerous factors about CHD.Methods Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) were used for the detection of MMP-9 and GPIIIa genotype on 64 CHD patients, including 37 myocardial infarction (MI) and 27 unstable angina (UA), and 84 healthy control subjects, and then the frequency of mutation was statistically computed. The level of plasma Fg , lipid ( TC, TG, LDL, HDL,) and lipoprotein (LPa, APOA, APOB) were measured by full-automatic biochemical apparatus.Results Three genotypes of MMP-9, C/C,C/T,C/T had been detected in both groups. The frequencies of C/T genotype and T allele in UAP were significantly higher than those in the control group(P<0.05), whereas the frequencies of GPIIIa C/T were very low both in CHD and control group. The level of plasma TG and Fg is higher, while APOA is lower in CHD than that in control group. These have showed significance difference(P<0.05). According to logistic regression analysis, APOA showed protectiveness to CHD while TG and Fg are dangerous.Conclusion MMP-9 C-1562T maybe an impressibility factor of UA below 65 years old in CHD; GPIIIaT1565C possibility is not a heredity factor of CHD in Shanxi; Abnormity of plasma lipid and coagulation system have involver in the development of CHD.
