Objective The aim of this study was to investigate the changes of plasma TXB2,6-keto-PGF1α and ET-1 in type 2 diabetic patients and their roles in the pathogenesis of diabetic macroangiopathy and to provide evidence for early diagnosis and treatment of diabetic macroangiopathy.Methods Plasma levels of TXB2,6-keto-PGF1α and ET-1 were measured with RIA in 60 cases of type 2 diabetic patients(43 patients with and 17 without diabetic macroangiopathy)and 31 cases of controls.Results 1.Plasma levels of TXB2 and ET-1 in type 2 diabetic patients were significantly higher than those in controls.Plasma levels of TXB2 and ET-1 in type 2 diabetic patients with macroangiopathy were significantly higher than those without macroangiopathy. However,plasma levels of 6-keto-PGF1α in type 2 diabetic patients were significantly lower than those in controls. Plasma levels of 6-keto-PGF1α in type 2 diabetic patients with macroangiopathy were significantly lower than those without macroangiopathy.2.Plasma levels of TXB2,6-keto-PGF1α and ET-1 had correlation with HDL-C,HbA1C,FPG;but didn't correlate with sex,age,BMI,SBP,DBP,CHOL and LDL-C.Triglycerides had positive correlation with plasma levels of ET-1,but didn't correlate with TXB2 and 6-keto-PGF1α.3.Plasma levels of TXB2 in type 2 diabetic patients had negtive correlation with 6-keto-PGF1α.Plasma levels of TXB2 in type 2 diabetic patients had positive correlation with ET-1.Plasma levels of 6-keto-PGF1α in type 2 diabetic patients had negtive correlation with ET-1.Conclusions 1.TXB2,6-keto-PGF1α and ET-1 in type 2 diabetic patients may play important roles together in the pathogenesis of diabetic macroangiopathy. Detemination of TXB2,6-keto-PGF1αand ET-1 levels in type 2 diabetic patients may be useful for early detection and serve as the index in monitoring the state of diabetic macroangiopathy.2.HDL-C,HbA1C and FPG may cooperate with TXB2,6-keto-PGF1αand ET-1 in the development of diabetic macroangiopathy.Strickly control the levels of plasma glucose and improve the level of HDL-C may postpone the development of diabetic macroangiopathy.3.High level of plasma TG had positive correlation with plasma levels of ET-1,which suggests that endothelial dysfunction existed.But whether high level of plasma TG play a role in the pathogenesis of diabetic macroangiopathy is needed for further study.
