Migraine is a kind of paroxysmal neurovascular dysfuntion, with character of repeated attack of headache in unilateral or bilateral head, it is often divided into the typical migraine, the common migraine and the special migraine. Migraine is very common in clinic, while the etiological factor of migraine is not clear, the mechanisms and pathophysiological process are also not understood completely up to now. Researches of many scholars have proved that prostaglandin(PG) is very important in the mechanisms of migraine. Thromboxane A2 (TXA2) and prostacyclin (PGI2) are a couple of compounds of PGs, which have been found in recent years. Thromboxane B2(TXB2) and 6-Keto-prostaglandin F1α (6-Keto-PGF1α), which are the stable products of TXA2 and PGI2, may reflect the content of TXA2 and PGI2 directly. There are a lot of common knowledge between the tranditional Chinese medicine and Western medicine in the migraine, Western medicines are not easily accepted because of its ill effect or expensive price , and the special advantages of traditional Chinese medicine have already been show in clinic, acupuncture therapy is just one of them.Objective To explore the content of plasma TXB2 and 6-Keto-PGF1 α in the jugular veins in models of migraine rats, and the effects of acupuncture on the content of plasma TXB2 and 6-Keto-PGF1α under the finished works, in order to get the further exploration of action mechanisms of the acupuncture to cure migraine.Methods The models of migraine rats were reproduced in accordance with Knynihar-Cslillik methods. The 40 rats were divided randomly into four groups ,each group has 10 rats: (1)Normal controlgroup, (2)Model control group, (3) Acupuncture control group, (4)Acupuncture treat group, then the rats were managed with scheme of each group. The blood in the jugular veins was taken after the experiments had finished. The content of plasma TXB2 and 6-Keto-PGF1α was detected by radio-immunity analysis method, with TXA2 and PGI2 concertration directly reflected.Results (1)The content of plasma TXB2 was constantly kept in higher level, and the content of plsama 6-Keto-PGF1α was constantly kept in lower level in the normal rats. (2)Model control group was in contrast with normal control group: the level of plasma TXB2 and TXB2/6-Keto-PGF1α decreased significantly (p<0.05), while the content of plasma 6-Keto-PGF1α increased significantly(p<0.01). (3)Acupuncture treat group was in contrast with model control group: the level of plasma TXB2 and TXB2/6-Keto-PGF1α increased significantly(p<0.05), while the content of plasma 6-Keto-PGF1α decreased significantly(p<0.05). (4)Acupuncture control group was in contrast with model control group: The content of plasma 6-Keto-PGF1α decreased significantly (p0.01), TXB2/6-Keto-PGF1α increase significantly(p<0.05), while the content of plasma TXB2 has no significal difference. (5)The level of plasma TXB2 , 6-Keto-PGF1α and TXB2/6-Keto-PGF1α in acupuncture control group has no significal difference compared with acupuncture treat group (p>0.05).Conclusions (1) The level of plasma TXB2 and TXB2/ 6-Keto-PGF1α decreased significantly, while the content of plasma 6-Keto-PGF1α increased significantly in the jugular veins in models of migraine rats may be one of mechanisms to trigger migraine, with aseptic inflammation developed by cerebral vessels dilatation. (2) After treatment withacupuncture, the level of plasma TXB2 and TXB2/6-Keto-PGF1α increased significantly, while the content of plasma 6-Keto-PGF1α decreased significantly in the jugular veins in models of migraine rats, regulating the synthesis and metabolism of plasma TXB2 and 6-Keto-PGF1α and maintaining the dynamic equilibrium of plasma TXB2 and 6-Keto-PGF1α may be one of mechanisms that acupucture can treat migraine effectively.
