Objective To explore the effect of Ravan Napas on serum soluble intercellular adhesion molecules-1 (sICAM-1) , the expression of ICAM-1 and HO-1 on bronchial epithelial cells and the eosinophils (EOS)in bronchoalveolar lavage fluid (BALF) of asthmatic rats. Methods The rats were divided into seven groups randomly: normal control group, asthmatic model group, model group treated with Ravan Napas three different dosages :high dosage, medium dosage and small dosage, model group treated with dexaraethason(DC) and the model group treated with Chinese traditional medicine. Rats were sensitized and challenged with ovalbumin to establish asthma model. The level of sICAM-1 in serum was measured by enzyme linked immunoabsorbent assay (ELISA), the expression of ICAM-1 and HO-1 on broncho -epithelial cells and the pathological changes in pulmonary tissue slides were detected through immuno-histochemistry and HE staining, the number of EOS in BALF were counted through direct counting method. Results The level of sICAM-1 in asthmatic group is significantly higher than the control group (P<0. 01), serum sICAM-1 level in Ravan Napas high dosage and medium dosage groups were significantly reduced after treatment (P<0.01); The expression of ICAM-1 and HO-1 on broncho -epithelial cells is significantly higher in asthmatic group than that of the control group(P<0.01), their expression was obviously inhibited in Ravan Napas high and medium dosagetreatment groups(P<0. 01);The number of EOS in BALF is greater in asthmatic group than normal control group(P<0. 01) and the number of EOS is significantly reduced in Ravan Napas high dosage group compared with the asthmatic model group(P<0. 01); There were more inflammatory cells infiltration in the tiny bronchial wall, hyperlasia of cup-shaped cells and thicker smooth muscle in the pulmonary tissue slides. The inflammatory cells infiltration was obviously decreased in model treated with Ravan Napas. Conclusion Ravan Napas can reduce the bronchial inflammation of asthmatic rats by regulating the amount of serum sICAM, expression of ICAM-1 and HO-1 on broncho-epithelial cells and the number of EOS in BALF of asthmatic rats.
