Objective: To study the efficacy and safety of KIO3 salt for treatment of iodine deficient goiter, we observed the treatment effect and antioxidative capability of KIO3 and KI in different dosage on goiter induced by iodine deficiency. Methods: Rats with iodine deficiency goiter were divided into 6 groups given by different dosage of KIO3 and KI for 3 months, respectively, that was normal iodine (1 fold), 5 fold and 50 fold KIO3 and KI. The other 2 groups of LI and NI remained as control. 1 month and 3 months later, the rats were sacrificed and some parameters were measured.Results: 1. (1) In 6 treated groups thyroid weights decreased markedly, but not completely recovered to normal. And the more iodine was given, the slower recover of goiter occurred. (2) (i) Urinary iodine was paralleled with iodine intake. (ii) Iodine content in thyroid of 6 treated groups was higher than LI group and lower than NI group, and no differences were found among 6 administrative groups. (3) Using routine histology methods and a MIAS-2000 image analysis system, the morphological observation and stereological analysis were performed. The results demonstrated that The mean follicular volume (V), mean surface (S) and volume density (Vv) in 6 administrative groups were significantly higher than that in LI group and lower than that in NI group. The numerical density (Nv) was lower in 6 administrative groups compared with the LI group, but still higher than that in NI group. Vv of the 50xKIO3 and 50XKI groups were significantly higher than that of the other 4 administrative groups. No differences were found between KIO3 and KI administrative groups at 3 iodine-supplying levels. The epithelial cells' volumes of 50xKIO3 and 50KI groups were lower than that of the other 4 administrative groups. At the same time, we found some large syncretic follicles in these two groups, but not in other 4 administrative groups. This may indicate theexisting of the cells' necrosis or apoptosis. (4) Serum TT4 in 6 treated groups was higher than LI and NI group, but no difference was found among 6 treated groups.After treatment for 1 month, level of serum TTa in the 1 xKIOs SxKIOs group waslower than LI group, but similar to NI group. After treatment for 3 months, there was no difference among 6 treated groups, LI and NI groups. (5) TT3 and TT4 levels in thyroid of 6 treated groups were higher than LI group. The levels of thyroid TLj in 1 fold and 5 fold groups were higher than NI group, and were higher than 50 fold administrative groups after 3 months' iodine supplement. No difference was found in thyroid TTs among 6 treated groups. (6) No differences were found between KIOs and KJ administrative groups at 3 iodine-supplied levels. (7) KIOs and KI are both effective on goiter involution. At the same iodine supplying level, no significance was found in all of the indicators measured in our experiments. So we can assure their treatment effects are the same. 2. (1) The capability of thyroid antioxdition: (i) The SOD and MDA in 6 treated groups were markedly lower than LI group and still higher than the NI group, (ii) After treatment for 1 month, the more iodine was given, the lower SOD and MDA were detected. Affter treatment for 3 month, the more KIOs was given, the higher SOD and MDA were detected, but no much change was found in 3 dosage KI groups, (iii) After treatment for 1 month, SOD of KIOs groups were higher than that of KI groups, but after treatment for 3 month, SOD of KIOs groups might lower than that of KI groups. MDA of KIOs groups might higher than that of KI groups at 1 and 3 month's treatment. (2) The capability of serum antioxdition: (i) There was no difference in SOD and MDA between LI and NI groups, (ii) SOD in the other 5 groups ,except 1 fold KI group, was similar to LI and NI groups. MDA of 3 different dosage KIOs groups were all lower than that of NI group. Furthermore, MDA of 1 fold KIOs group was also lower than that of LI group.
