Objective: To study effect of myocardial protection perfused by bone marrow cells on ischemia-reperfusion injury in isolated rat hearts and its machanism. Methods: On modified Langendorff apparatus, 24 isolated SD rat hearts were randomly divided into 3 groups (n=8): I/R group (hearts suffered from 40-minute global anoxia /40-minute reoxygenation); bFGF group (previously hearts were perfused with oxygenated perfusate administered with 8μg bFGF, then subjected to 40-minute global anoxia/40-minute reoxygenation); BM-MNC group (previously, hearts were perfused with oxygenated perfusate administered with 1.5-2.0×106/ml BM-MNC before it suffered from 40-minute global anoxia /40-minute reoxygenation)。The hemodynamic parameters and leakage myocardial creatine (CK) were measured before ischemia and during reperfuion. Myocardial activity of superoxide dismutase (SOD),Na+-K+-ATPase,Mg++-ATPase,Ca++-ATPase and malondialdehyde(MDA) content were examined. Myocardial ultrastructure were observed. Results: Compared with I/R group, BM-MNC group or bFGF group increased coronary flow, improved heart function; notably promoted activity of superoxide dismutase (SOD),reduced malondialdehyde (MDA) content and myocardial creatine; furthered activity of Na+-K+-ATPase, Mg ++-ATPase and Ca++-ATPase in myocardium. Myocardial ultrostructural changes: the myocardial ultrostucture in I/R group was damaged,which characteristized by mitochondrial swelling and crack, disappearance of mitochondrial crista, disarrangement of sarcomere and/or disruption. As compared with I/R group, the changes in BM-MNC group or bFGF group was significantly better, with only slight swelling of sarcomere and mitochondrion.Conclusions: Experimens showed that BM-MNC group may reduce myocardial ischemia - reperfuion injuries in the isolated rat heart...
