Abstract/Summary:
ObjectiveAt present, incidence rate of cervical cancer is the second highest of the female malignant tumor. About 500 thousand new cases are reported in the world every year, almost 5 percent of the incidence rate of all cancers. Before effective vaccine of human papillomavirus ( HPV) have been applied, screen for cervical cancer is the main way to prevent and control cervical cancer. Screen of cytology contribute most to reduce the incidence rate and mortality of cervical cancer. As the studies of etiology, high - risk HPV is significantly relative to the precursor lesions for cervical cancer - cervical intraepithelial neoplasia (CIN) and cervical cancer. High - risk HPV is the necessary biologic cause of the precursor lesions of cervical cancer and cervical cancer. And with the rate of detection increasing, diseases are more serious. It can be inferred that high -risk HPV can predict the risk of infected women. With the improvement of detectable way, it is an important part of screen for cervical cancer. This study discusses the infection of high - risk HPV expresses in normal cervix, benign lesion, CIN and cervical cancer comparing with thin - layer cytology test, and evaluates the function that can improve the positive ratio of CIN and cervical cancer by the high - risk HPV test methods.Methods1. Subjectives:The clients came from outpatient of liaoning Cancer Hospital from January 2004 to May 2005, with their cervical diseases confirmed by colposcopy. Totally 357 cases.2. Test HPV DNA with hybrid capture method:Hybrid capture II HPV DNA system and test box for high - risk HPV are supplied by Digene corporation. Probe comprises of 13 subtypes, type 16, 18, 31, 33, 35, 39, 45, 51, 52, 56, 58, 59, 68. The main steps are (1) spall-ation (2) hybridization ( 3 ) capture (4) response (5 ) flush (6) collecting sign (7) outcome judgment; the content of HPV DNA is above lpg/ml, which means the ratio between RLU and CO is above 1, positive.3. Thinprep cytologic test;Cells from cervix is collected with the special brush , the specimens are conserved in a bottle with preservative fluid. After making thin - layer, being fixed by 95% alcohol, pap staining, diagnosing by pathophysiological doctors, the specimen classified with ASCUS and above are positive.4. Pathology by colposcopy:Bishops from aceto - white epithelium, mosaic, white glade, atypical vessels and 4 points of transformation zone from normal women are sent to checked for the last diagnosis.5. Stastical analysis:X~2 test is applied (a = 0.05) and software SPSS 11.5 is used for data a-nalysis.Outcome1. The ratio of high - risk HPV detected from CIN I group is 49.1% ,CIN II group is 57.9% , CIN HI group is 92.5% , and cervical cancer group is 98.3%. There is significant difference between test group and controll group (P < 0.05). In conclusion, infection of high - risk HPV is relative to occurrence of cervical cancer.2.
