The orthogonal design was use to optimize softening and cutting process of Radix Bupleuri with the content of total saponin and essential as markers. I have used HPLC for determination the content of saiko saponin a and d in Radix Bupleuri .1 have the Conclusion that saiko saponin a were linear in range of 0.275μg~1.925μg (r=0.9997); saiko saponin d were linear in the range of 0.250μg~1.750μg (r=0.9998) .This method is simple and reproducible and can be used for the quality control of Radix Bupleuri .To study the quality of Radix Bupleuri .the quantity of inorganic matter, water content and content of saiko saponin a and d in Radix Bupleuri were analyzed, saiko saponin a and d in Radix Bupleuri were identified by TLC. The quantity of saiko saponin a and d in Radix Bupleuri were assayed respectively in 0, 1, 2, 3, 6, 12 months. The results indicated Radix Bupleuri had stable component quantity in one year .This study of Radix Bupleuri have been confirmed standard and will be used in market and industrial production.To establish the method of fingerprint analysis of Radix Bupleuri from Hebei by HPLC. The fingerprint consists of 13 common peaks .I have the Conclusion that the method is accurate , reliable and can be used in quality evaluation of Radix Bupleuri from Hebei.
