Objective : At the present time, the research of the platelet activation is focused on thecorrelation in the internal medical diseases, such as cardiac or cerebrovascular diseases, diabetes mellitus and inflammatory diseases. But the research of the platelet activation, especially platelet microparticle , is rare in patients with arteriosclerosis obliterans of legs accompanied by essential hypertension(LEAO EH). To investigate the change of platelet activity in patients with LEAO EH and determine whether platelet is activiated in patiens with LEAO EH, we measured the expression of membrane glycoproteins(GP) on platelet microparticles and the level of platelet aggregation rate (PAgT) in patients with EH and with LEAO EH (n=20), the elder essential hypertensive patients (n=20) and healthyvolunteers(n=20) served as control. Methods: The platelet activation markers includingmicroparticle membrane glycoproteins of CD62p PAC-1 were fluorescence labeled with their corresponding monoclonal antibodies and analyzed by FCM, and the change of theplatelet aggregation rate (PAgT) was investigated together in all patients. Results : 1.Of three group, expression level of CD62p PAC-1 in EH group and LEAO EH group is increased. On the one hand, the expression of CD62P PAC-1 is higher in LEAO EH group than in pure EH group. 2. Platelet aggregation rates in patients of EH and LEAO EH are increased significantly;there was more increase in EH and LEAO EH group than that in pure EH group. Moreover, the expression level of CD62P PAC-1 was positively correlated with the platelet aggregation rate (r=0.8701, 0.7931, p<0.01)Conclusion: 1. CD62p PAC-1 , The platelet microparticle activation markers,reflected the change of platelet activity better. 2.The expression of CD62p PAC-1 and PAgT is higher in the pure EH group and LEAO EH group, and the increase of LEAO EH is more obvious. These results suggest that increase of platelet activity and platelet aggregation rate are related to occurrence of LEAO EH.
