Object: To detect the positive rate of HPV16 E6 gene in esophageal cancer tissues and study the structural specificity of HPV16 E6 gene from kazakh's esophageal cancer tissues of Xinjing patient. Methods: HPV16 E6 gene of esophageal cancer was amplified by PCR technique, investigating the relationship between HPV16 E6 and esophageal cancer. based on the research object from kazakh's esophageal cancer tissues, Cloning and sequencing of HPV16 E6 gene from kazakh's esophageal cancer tissues. Results: 8 cases and 16 cases HPV16 E6-positive sample were deteced in tumor-adjacent normal tissues and esophageal cancer tissues. 8 cases HPV16 E6-positive sample of eighteen cases kazakh's esophageal cancer tissues from Xinjing patient, and the positive rate was 44.44%. HPV16 E6 gene of kazakh's esophageal cancer was successfully cloned. In comparison with the prototype accepted by GeneBank, the deduced amino acid sequence of HPV16 E6 protein was changed. Conclusion: Variations take place in the nucleotide sequence of HPV16 E6 gene obtained from kazakh's esophageal cancer tissues of Xinjing patient, there were one point mutation in HPV16 E6 nucleotide sequence from A to G in 28 base pair and the deduced amino acid Sequence of HPV16 E6 protein was changed from Arg to Gly. induce the E6 protein to degrade p53 function and other immunogenicity were changed.
