Objective: Blood stasis can be found in various clinical disease sections. Microcirculation disturbance is the basic pathological changes of blood stasis. There are close relations and similar laws between activating blood circulation to dissipate blood stasis and improving microcirculation. In mechanisms of microcirculation disturbance, platelets hold the important status. Platelets activation and hyperfunction play the vital role in blood stasis. This experiment wants to study the relevance of platelet functional elements and blood stasis andinvestigate the mechanisms of the water-soluble preparation of Danshen(radix salviae miltiorrhizae) Danshen Injection on the disturbance of mesentery microcirculation in rats and platelet functions in rats with blood stasis. Methods: Experiment One: Effects of Danshen Injection on mesentery microcirculation inrats: Rats were divided into 5 groups, such as microcirculation disturbance model group, Danshen Injection group (high dose, middle dose, low dose), andanisodamine (654-2) group. Medicines were intragastric administered to the rats consecutively for five days. On the fifth day, the abdomen of the rats was opened 30 minutes after medicine administration and observed diameter of mesentery arteriole, blood velocity, density of capillary and the time of the bloodstream recovering. Experiment Two: Effects of Danshen Injection on platelet functions in rats with blood stasis: Rats were divided into 6 groups, such as normal group, blood stasis model group, Danshen Injection group (high dose,middle dose, low dose ), and aspirin group. Medicines were intragastric administered to the rats consecutively for five days. On the fourth day, the blood stasis model was induced with adrenaline and ice stimulation 60 minutes after the Medicines administration. On the fifth day, the blood of rats was collected 60 minutes after the Medicines administration to evaluate platelet function. The plasma level of PGI2 and the platelet level of TXA2, cAMP were determined by radio-immune assay. The platelet level of GMP-140 was measured by competetiveenzyme-immuno assay. Results: Experiment One (1) Compared arteriole diameter with before challenge, every group decreased at lmin, 3min, 5min,7min after giving adrenaline (P<0.01);At 9min, Danshen Injection high dose group and anisodamine group have recovered to the level before challenge. Compared with microcirculation disturbance model group, at lmin,3min,5min,7min,9min anisodamine group and Danshen Injection high dose group had significant difference (i><0.01) (P<0.05);At 9min, Danshen Injection middle dose had significant difference ( P<0.05).(2) Compared blood velocity with before challenge, every group decreased at lmin, 3min,5min,7min after giving adrenaline (P<0.01);At 9min, Danshen Injection high dose group and anisodamine group have recoverd to the level before challenge. Compared with model group, on 5 time point, anisodamine group had significant difference (P<0.01)(P<0.05);At 3min, 5min, 7min, 9min, Danshen Injection high dose group had significant difference (P<0.01) (P<0.05);At 5min,9min, Danshen Injection middle dose group had significant difference (P<0.0l). (3) Compared density of capillary with before challenge, every group decreased at lmin, 9min after giving adrenaline (P<0.0\). Compared with model group, there wasn't difference among all groups at lmin. At 9min, anisodamine group and Danshen Injection high dose group had significant difference(P<0.01).(4)Compared the time of the blood stream recovering with model group, anisodamine group and Danshen Injection high dose group had significantly difference (P<0.01). Experiment Two: (1) Compared the platelet level of TXA2 with blood stasis model group, every group decreased (P<0.01). (2) Compared the plasma level of PGI2 with blood stasis model group, Danshen Injection low dose group increased (P<0.05), other groups increased (P<0.01). (3) Compared the level of TXA2/ PGI2 with blood stasis model group, Danshen Injection low group decreased (P<0.05), other groups decreased (P<0.01). (4) Compared the platelet level of cAMP with blood stasis model group, Except Danshen Injection low group, other groups increased (P<0.01). (5) Compared the platelet level of GMP-140 with blood stasis model group, Danshen Injection middle, high dose group decreased (P<0.01). Conclusions: (1) The modelsof two experiments were established successfully. (2) Danshen Injection has a good effect on mesentery microcirculation disturbance in rats. (3) Danshen Injection has a dramatic effect on rats of blood stasis through decreasing the platelet level of TXA2, GMP-140 and the level of TXA2/ PGI2;Increased the plasma level of PGI2;and the platelet level of c AMP.
