Diabetes is one of the three chronic non-communicable diseases that threaten human health.The body is in a state of hyperglycemia for a long time,often causing multiple complications.Diabetes is plaguing people's health and affects people's quality of life.As an effective drug for the treatment of diabetes,insulin is often used in combination with traditional Chinese medicine injections for the treatment of diabetic complications.Danshen Injection has the effect of promoting blood circulation,removing blood stasis and nourishing the heart.Usually,the free state of the drug has biological activities,and the binding state affects its efficacy in the body.Therefore,whether the combination of drugs and insulin will affect the efficacy of drugs is of great significance for guiding clinical drugs.Objective: Study whether the combination of insulin and the main active ingredients of Salvia miltiorrhiza injection will affect the drug activity.,and discuss the optimal drug delivery way of insulin and traditional Chinese medicine injection.Methods: Determination of the combination situation of insulin to salvianolic acid A or salvianolic acid B by electrospray ion trap mass spectrometry.Rat model of blood stasis syndrome of type ? diabetes was induced by high fat/high sucrose diets and streptozotocin united prednisolone and epinephrine.Single channel intravenous drip was adopted with insulin and Danshen Injection mixture,and double channel with insulin and Danshen Injection separately.Blood plasma were collected by abdominal aortic method to measure fasting blood glucose level,serum insulin value,serum malondialdehyde and blood rheology indexes.The kidneys and abdominal aorta of the rats were selected to make frozen sections.Determine the better mode of drug delivery by comparing the differences between the various indicators.Results: According to the mass spectrum,insulin and salvianolic acid A,as well as insulin and salvianolic acid B,all produced non-covalent complexes,and the binding ratio was 1.After modeling,In the type II diabetes blood stasis syndrome group,the hair color of the rats was dry and dull,the weight was reduced,the mental state was poor,and the diet and urine volume increased.The lumen of the renal tubules becomes larger and dilated,the intimal lesion of the abdominal aorta is broken,and the inner surface of the lumen is not smooth.After treatment,the hair color of the rats was slightly shiny and the mental state was good.In the double-channel group,the color of the tongue was lighter than that in the single-channel group,the auricle was ruddy,and the renal tubule adhesion decreased.Compared with the single-channel group of rats,the intimal injury of the abdominal aorta in the double-channel group was significantly improved,and there was a partial intact wavy intima;And compared with the normal control group,the intima-media thickness of the abdominal aorta in the model control group was significantly different(P<0.01).Compared with the model control group,the abdominal aorta intima-media thickness was significantly different between the double-channel group(P<0.01),and there was no statistically significant in single-channel group(P>0.05).Compared with the normal control group,the low-cut and middle-cut in the diabetic blood stasis syndrome group were different(P<0.05),the high-cut and erythrocyte aggregation index were significantly different(P<0.01).Compared with single-channel group,the low-cut and the high-cut in the double-channel group was significantly different(P<0.01),and the middle-cut and erythrocyte aggregation index were no difference(P>0.05).Compared with model control group the middle-cut in double-channel group was different(P<0.05),but the middle-cut in single-channel group was no difference(P>0.05).Compared with normal control group,the erythrocyte aggregation index in double-channel group was no different(P>0.05),but the erythrocyte aggregation index in double-channel group was significantly different(P<0.01).Compared with the normal control group,the insulin resistance index in the diabetic blood stasis syndrome group was significantly different(P<0.05);Compared with the single-channel group,the insulin resistance index in the double-channel group was different(P<0.05).Compared with the single channel group,the serum malondialdehyde in double-channel was significantly different(P<0.01).Through the HE staining pictures of the abdominal aorta and kidney and the comparison of blood rheology indexes,the improvement effect of the double-channel group is more obvious than that of the single-channel group.The kit experiments showed that the double channel group serum insulin resistance index and serum malondialdehyde improvement were more obvious.Conclusions: Insulin combines with the active ingredients of Danshen Injection,so that the drug can not work better.We can see that the effect of the double-channel group is obviously superior to that of the single-channel group.Insulin and Danshen Injection mixture would affect the efficacy of Danshen Injection.
