The Effect Of Danshen Injection On The Platelet Function In The Normal And Blood Stasis Rats

Objective:The main physiologic functions of platelets are haemostasis and thrombosis, and platelets play a vital role in atherosclerosis, the hyperplasy of smooth muscle and inflammation. When vascular endothelium gets injured, the flow of blood changes, or as a result of simulation of chemicals, a series of related changes occur in platelets, such as the change of shape, adhesion, aggregation and release reaction, which consequently facilitate the process of thrombosis. In order to investigate the mechanism of the water-soluble preparation of Danshen(radix salviae miltiorrhizae) on activating blood circulation to dissipate blood stasis, we tudy the effect of Danshen injection on the platelet function in normal and blood stasis rats. Methods: Experiment One The effect of Danshen injection on the platelet function in blood stasis rats: Rats were divided randomly into 6 groups, such as Normal group, model of blood stasis group, Danshen injection group [high dose (8.1g/kg), middle dose (2.7g/kg), low dose(0.9g/kg)], and aspirin group(4.0mg/kg).Medication was administered to every rat consecutively for five days. On the forth day, 60 minutes after the medication, the blood stasis model was established with adrenaline and ice stimulation. On the fifth day, 60 minutes after the medication, the blood of rats was collected to determine platelet function. The colorimetric method in microtiter plates was used to measure platelet adhesion, turbidity method was used to measure platlet aggregation, and the competitive enzyme immunoassay was adopted to determine the level of glycoprotein (GP) II b and IIIa; Experiment Two: HE staining was used to observe the changes of pulmonary morphology ; Experiment Three The effect of Danshen injection on cytosolic free calcium concentration([Ca²⁺]_i) in normal rat platelets.
