Shamoga is a common traditional Chinese medicine which comes from the branches and leaves of Artemisia halodendron Turcz. According to clinical investigation, Shamoga has many pharmacologic functions including eliminating phlegm, restraining asthma and expelling rheumatism and so on. It has been used for the treatment of chronic bronchitis, the rheum and the rheumatoid arthritis. It has not been embodied in Pharmacopoeia of People's Republic of China (2005 edition). Its main officinal part is branches and leaves refering to literature, but seeds by clinical investigation and folk use.Heretofore, the report on the active ingredients and pharmacological activity of Shamoga is scarce. The chemical constituents and anti-inflammation pharmacodynamics have been researched by our group. Based on the previous study, we take eliminating phlegm and restraining asthma as pharmacology target in the study. The effective parts of Shamoga seeds were screened, the active ingredients pursued, its effective ingredients found and its pharmacology functions nailed down. So the primary quality control methods were built. And pharmaceutical bases were supplied for its use and exploitation as a new drug. Main researches are as follows:1. Pharmacodynamic screening: To study the anti-asthmatic effective part of the Shamoga seeds, we tested the action in the three different models-eliminating phlegm, immediate asthma reaction(IAR), late asthma reaction(LAR), using three animal tests(Phenol red eliminating phlegm in mice, histamine spray to asthma in guinea pig, OVA antigen induce allergic airway reaction in mice). Meanwhile, effective parts were screened with traditional polarity sort method. As a result, Shamoga seeds were effective on eliminating phlegm and late asthma reaction(LAR) and the effective parts were found to be part B and part C.2. Components study: To research chemical components of the anti-asthmatic effective part, part C was extracted and isolated, and five compounds were received consequently. By chemical and spectroscopic analysis , the five compounds were identified as isosakuranetin(Ⅰ) , 4′,7-dimethylkaempferol(Ⅱ), eriodictyol-7-methylether(Ⅲ), Rhamnetin(Ⅳ), daucostorol(Ⅴ) .ⅠandⅡwere found in this plant for the first time. Part B of Shamoga seeds was analyzed by GC-MS. 34 compounds were isolated and identified, most of them were unsaturated fatty acid and terpene, in which the content 9,12-Octadecadienoic acid ethyl ester was the most. In addition, there were 19 kinds of compounds being the same as those in branches-leaves.3. Screening of 5-LOX anti-inflammation model in vitro: To study active ingredients and targets of anti-asthmatic effective part of Shamoga seeds, effective parts and active...
