| It is widely recognized that Podophyllum plays multiple roles in pharmacological and biological function as a traditional Chinese herbal medicine. This paper is performed on extraction technology, purification and immune function of Podophyllum Emodi polysaccharides(PEP). The main results are as follows:1 Extraction technology and components of PEP500g Podophyllum was degreased,eliminated pigment after being desiccated. powder of Podophyllum was extracted for hours in hot water according to designed solid-liquid ratio, temperature, time. Then, liquid was dealt with ethanol,acetone,aether respectively. Finally, 31.9g of Podophyllum Emodi polysaccharides without amylum or protein was obtained. The polysaccharides especial apices were seen by the infrared spectrum scan: 3412.47cm-1,2928.50cm-1,1648.25cm-1,1560.28cm-1,1368.97~1456.81cm-1,929.70cm-1 and 850.37cm-1;It was proved by Thin-layer chromatograms that PEP was composed of glucose, fructose, mannose and arabinose.2 Function of immune adjustmentThe investigations on immune functions of PEP mainly include: spleen index, thymus index, content of bacteriolysin in serum, lymphocytes proliferation, gene expression and the effect on reactive oxygen species(ROS) free radicel.2.1 Effect of PEP on spleen index, thymus index and content of bacteriolysin in serumOn Day 1,3,5,7, PEP(200mg/kg) treated to mouse by ig can improve the spleen index, difference(P<0.05)compared with control was observed. But the polysaccharide has no effect on the thymus index or the content of bacteriolysin in serum. On Day 1~7 ,PEP(200mg/kg) treated to mouse by ip can improve spleen index obviously, there were significant difference(P<0.01)compared with control.2.2 Effect of PEP on lymphocytes proliferationThe lymphocytes isolated from murine spleen were cultured according to different concentrations of PEP. After 3 days, MTT method was used to investigate the effect of PEP on lymphocytes proliferation, the result showsed PEP at different dose had different effects on lymphocytes proliferation. 50μg/ml PEP had obviously effect, 100μg/ml,200μg/ml,400μg/ml PEP had more effect. PEP was able to induce the lymphocytes proliferation in cooperation with ConA. It also can stimulate the lymphocytes proliferation when hydrogen peroxide was added to culture medium .2.3 Effect of PEP on gene expression... |
