Objective:To explore the antitumor effect of Tagalsin (extracted from Ceriops tagal) and it’s immunomodulatory effect on transplanted subcutaneous sarcoma in H22and Ehrlich ascites tumor cells (EAC) mice, moreover, to reveal the inhibitory mechanism of Tagalsin.Methods:Animal models were established by inoculating H22mouse hepatoma cells and Ehrlich ascites tumor cells to the skin of left armpit in mice, and then were randomly divided into5different groups:the high dose of Tagalsin (16.50mg/kg· d), the middle(3.30mg/kg· d), and the low(0.66mg/kg· d), equal volume of edible oil was applied to control group(negative control),and Carmofur Tablets (100mg/kg· d) was used as positive control, All the groups of mice gavaged with the above solutions for10days, each group of mice was observed for one day after the last administration, and then all the mice were killed after weighting the qualities, count the body weight changes, and the tumor weight to calculate the rate of tumor restrain respectively, the spleen and thymus indexes were evaluated according to spleen and thymus weights respectively. The expression levels of bcl-2/bax were detected by immunohistochemistry, the apoptotic index(AI) was determinated by the terminal deoxynucleotidyl transferase-mediated nick end labeling method(TUNEL).Results:The body weights among mice were not statistically different before experiment(P>0.05), the body weights in high or mid concentrstion Tagalsin group increased statistically higher than that in control group after experiment; Tagalsin can inhibit the growth of tumor, compared with control group, the tumor grew relatively more slowly in positive control group and doses of Tagalsin groups (p<0.05); Compared with control group, positive control and doses of Tagalsin decreased the indexes of spleen and thymus evidently (p<0.05), compared with positive group, the index of thymus in all doses of Tagalsin were lower than the positive group, and the difference was significant (p<0.05); Immunohistochemical analysis showed that the ratio of bcl-2/bax in groups of different doses of Tagalsin and positive control had statistical difference compared with control group (p<0.05); The apoptotic indexes(AI) of treatment group in tumor-bearing mice were higher than control group, and the highest in positive control group, and had statistical difference.Conclusion:Ceriops tagal extracts Tagalsin can inhibit tumor-bearing mice of H22and EAC more or less, and showed a weaker humoral immunosuppressive in mice. Tagalsin inhibits the growth of tumor in mice to achieve its inhibitory mechanism by decreasing the protein expression of bcl-2, and increasing the expression of bax, and reduce the ratio of bcl-2/bax, and then promote tumor cells toward apoptosis.
