To further study the expression level of leptin in the alcoholic fatty liver disease and the mechamsion on this disease, we could establish the model of alcoholic fatty liver in rats fed alcohol by a physiological feeding way in this study. So some enzymes were determined from plasma and liver homogenate by using the methods of spectrophotometry and ELISA. The expression levels of leptin mRNA and protein were determined by RT-PCR and immunohistochemisty. The results showed:1. In the model of alcoholic fatty liver in rats fed alcohol by a physiological feeding way, the results on liver histology were analyzed: The livers in alcohol group were engorged with the fatty deposition and different inflammation and putrescence. The hepatocytes of rats in alcohol group presented the diffusion steatosis, and in the local part inflammation and putrescence. And compared with normal group and control group, the liver coefficients in alcohol group were higher, which further showed that the model of alcoholic fatty liver in rats was established.2. Some relative enzymes (ALT, AST) in plasma and the levels of TNF-аand leptin were determined by using the methods of the spectrophotometry and ELISA. The results showed: the levels of ALT, AST, TNF-аand leptin were higher than those of control group and normal group.3. GSH and SOD activity and MDA concentration from 10% liver homogenate were determined by using the 722 UV-visible spectrophotometer. The results showed: compared with normal group and control group, GSH and SOD activity in alcohol group was lower, but MDA concentration was higher.4. The expression levels of leptin protein from liver tissue were determined by using the immunohistochemisty in alcohol group. The result showed: the positive was showed in Kuffer cells in alcohol group, which was mainly expressed in the core. And the positive was also expressed in a few amount of hepatcytes and hepatic stellate cells.5. The expression levels of leptin mRNA were determined by using RT-PCR in liver tissue. The results showed: the expression levels of leptin mRNA were increased in alcohol group, and the ratio of leptin and B-actin was also higher than the control group and normal group.We established the model of alcoholic fatty liver in rats fed alcohol by a physiological feeding way, which could study the expression level of leptin and the correlation with leptin and some relative enzymes in this model. And the effect of leptin was showed in the alcoholic fatty liver disease. At the same time, these results would provide the good basis in alcoholic fatty liver disease and therapeutic agent.
