Objective and Background:NF-Kappa B ( NF-κB ) has been one of the most hot research points since 1986 when it was found. NF-kB participates in not only lots of physiology regulation such as immune, growth and development, maturation and apoptosis of cells, but also many pathologic courses including inflammation, rheumatic diseases, cardiovascular diseases, neural diseases and cancer, et al. Epidermal growth factor receptor (EGFR) locates in membrane of many kinds of epithelia, influencing cells growth, proliferation and differentiation. Helicobacter pylori, one kind of gram-negative curve bacilli identified in 1983, isolated from different chronic gastric diseases and had been categorized as group I carcinogen for gastric carcinoma by World Health Organization. Years of research work have been done to reveal the relationship between H. pylori and gastric carcinoma. So far the correlation between H. pylori and gastric carcinoma mostly depends on epidemiology data and what's the exactly pathogenesis is still unknown. NF-kB or EGFR signaling pathway is activated abnormally in many tumors, hence they are all suspected to be involved in malignant transformation of tissues, carcinoma cells invasion and metastasis. Chaoshan District, located in South China, own special diet and has high prevalence of gastric carcinoma. Through investigating NF-kB and EGFR expression in mucosa epithelia of H. pylori-associated gastric precancerous lesions and carcinoma, we explore and discuss the possible pathogenesis of H. pylori-associated gastric carcinoma in the eastern part of Guangdong Province, South China. Materials and Methods:Total 152 cases of gastric carcinoma patients who came from the eastern part of Guangdong Province, were investigated by us (between March 2004 and April 2005 ). They all underwent H. pylori detection before surgery by rapid urease test, 14C-urea breath test, modified Giemsa stain, bacterial culture, finally 32 cases with H. pylori infection were enrolled in molecular biology research. All specimens were gained from patients underwent complete surgical resection including carcinoma tissue, tissue adjacent to the carcinoma and normal mucosa far from cancerous lump. Nuclear protein in mucosa extraction was stored at -80℃ after measured its concentration. Western blot was used to analyze NF-κB in nuclei, NF-κB DNA binding activity was determined by EMSA.
