Experimental Study On Treating Helicobacter Pylori Associated Gastritis With The Method Of The Integration Of Chinese Herbal And Western Medicine

Aim To screen mouse-adapted human Helicobacter pylori(Hp) strain developed chronic gastritis and to establish Helicobacter.pylori associated gastritis(HpAG) mouse model with BALB/c mouse we investigate the bacteriostasis in vitro and eradication therapy of Hp in vivo with Chinese and western medicine hoping the study can provide ideal drug against Hp for clinician.Methods 1. We collected samples of gastric mucosa from patients' stomach who live in our region and suffer from diseases of digestive tractand and Helicobacter pylori were isolated from those patients.2. After the bacteria grown, they were identified by gram staining and biochemical tests of urease, catalase and oxidase. The strains were confirmed as cagA~+ and vacA~+ by PCR. The bacterial inoculum was adjusted to a concentration of approximately 10~9organisms/ml and immediately used for inoculation by orogastric gavage to establish the model of Helicobacter pylori associated gastritis in BALB/c mice. 3. Drug sensitivity test was done in vitro by using Chinese herbal medicine, Western medicine and the combination of these two drug respectively.4. Based on the results of sensitivity test, effective and synergism drugs were used to treat the mice model by gavage.Effective of these drug in different groups were surveyed in the both of gastric inflammation and eradication of H.pylori.Results 1. We isolated 54 strains from 115 patients' gastric biopsies and of these 54 strains there were 32 cagA~+ and vacA+. 2. One of these 32 strains which can permanant colonize in the stomach of BALB/c mice were screened. 3. Dahuang,Huanglian,Huangbai, Huangqin and Huangqi were determined to be hypersensitive . Baiji was low sensitive. These Chinese herbal medicine were synergism . In western medicine, omeprazole, Clarithromycin and amoxicillin were all synergism each other. 4. Results of drug action shows that the group of mice treated with herb and western medicine were better than that of treated with herb or western medicine alone in improvement of gastric inflammation and eradication of Helicobacter.pylori.Conclusion 1. Helicobacter.pylori associated gastritis mice model was established with mice-adapted strain in our study .This model can be used to evaluate the action of drugs.2. It was determined that using both herbal and western medicine is better than using themalone in improvement of gastric inflammation and eradication of H.pylori and this therapywas worth being spreaded in clinic.
