In the paper, the Ganoderma Lucidum powder was used as research material and the Ganoderma Lucidum polysaccharide was explored in the following respects, including extraction, content determination, gradation, purification ,structure identification and activity. The results were as follows:The results showed when the temperature was at 90 ℃ , the ratio of material and water was defined as 1:15, the heating duration was 1.5h ,then centigraded ,the ratio of material and water was defined as 1:10,the heating duration was 1h, the polysaccharide extraction rate was 1.02%; The precipitating polysaccharide of 40%,65%,80% ethanol concentration contained the rate of 0.31:0.27:0.42.The 3 grades crude polysaccharide were soluted in 0.1mol/L NaCl solution at a flow rate of lml/2min with DEAE-52 ion-exchange column chromatography.The polysaccharide was detected and collected with phenol-sulphuric acid method.The elution profile of the GLP on DEAE-52 ion-exchange column was eluted with Sephadex G-75 column for the further purification in 0.06mol/L NaCl solution and 0.15 mol/L PBS(PH7.2) at a flow rate of 1ml/5min ,and then collected four concentration.Its monose composition was analyzed by acid hydrolysis and GC. It was composed of L-arabinose, D-galactose , D-glucose, L-rhamnose, D-mannose and L-fucose,and different ethanol gradation sediment has different monose composition .Its structures were studied by infrared spectra and Ultra-Violet spectrophotometry. GLP3 , GLP4 existed as β -pyranose and contained little protein and glucurronic acid. The weight-average molecula mass of GLP1,GLP3 and GLP4 were 4.89×10~4,2.14×10~4 and 0.99×10~4 respectively.
