ForewordThe integrity of sperm functions—sperm quality is the base of the normal fertilization and embryo development. In assistant reproductive technology (ART), whether we can choose the functional sperm to fertilize and use the safe sperm preparation technique, not only correlated to the outcome of ART, but also correlated to the safety of generation. Routine sperm analysis is used widely in evaluating the sperm quality, but limited in predicting the fertilization ability of sperm, and has visible differences between laboratories; And also the morphology and motility of sperm is not alwals consistent with the integrity of sperm's nucleu? DNA (nDNA), with the help of man-made choosing sperm of good morphology and molility when using micro-intracytoplasmic sperm injection (ICSI) can not insure the normality of sperm's germ plasm. So it is necessary to found more objective methods in clinical practice to evaluate the quality of separated sperm, and help the patients to choosing the reasonable treatment.ObjectivesTo find out more objective index to evaluate the sperm quality, and combine the routine sperm analysis parameters to evaluate a new cryopreservation method—cryopreservation of the prepared sperm; to find out objective index to evaluate the sperm DNA integrity, and explore the relationship between sperm DNA integrity and male inferlility.① Using normal prepared technique in clinical practice, and selecting specifical sperm colony, trying to investigate the interrelationship between early apoptosis, mitochondrial membrane potential(△ψm), reactive oxygen specials(ROS) of selecting sperm and the routine sperm parameters, and choosing the related indexes as the objective standard of sperm quality in this study.②To investigate effects of preparing technique in sperm cryopreservation on routine sperm parameters, mitochondrial membrane potential(△ψm) and reactive oxygen specials(ROS).③To investigate the integrity of nDNA in male infertile patients, and the effect of...
