Objective: To investigate P504S CK34βE12 as a new tumor marker for diagnosing prostate cancer (Pca). To provide experimental evidence and theory guidance for the diagnosis of prostate cancer. Methods:1. 116 prostate specimens, including 37 prostate cancers confirmingby needle biopsies, 8 prostate cancers confirming by and 56 BPH 15 high grade PIN were retrospectively analyzed. All the samples were measured for P504S CK34βE12 by immunohistochemistry.2. Analyse the results of immunohistochemistry and the relationship to Gleason, PSA and pathological stage. To find a way to the later treatment of the prostate cancer. Results:1. 91.1% of prostate cancer in our groups had P504S positive response, while all BPH were negative response.2. We made three groups depend on Gleason score .The first group 2-4 scores ,the second group 5-6 scores, the third group 7-10 scores.Higher score higher level of P504s. The expression of P504S was different between three groups (0.01

0.05)4. And the expression of P504S has relationship with pathological stage (p< 0.05).Conclusions:1. P504S can help urologists and pathologists to confirm the diagnosis of Pca, especially in small needle biopsy specimen.2. P504S also can be a new approach for the etiology and treatment research of prostate cancer.
