Objectives To assess the value of P504S/CK34βE12/p63 double-chromogen reaction for the diagnosis of prostate adenocarcinoma.To find different serum protein expression maps in prostate cancer and BPH by using surface enhanced laser desorption/ionization time of flight mass spectrometry(SELDI-TOF-MS).To find the biomarkers in prostate cancer for early stage diagnosis.Methods Using immunohistochemical SP and cocktail methods to investigate the expression of P504S/CK34βE12/p63 of prostate neoplasms,and to make diagnosis.Using SELDI-TOF-MS to analyze serum of these cases on WCX2 protein chip.The data was analyzed by biomarker wizard software.Results Sensitivity and specificity of triple-antibody P504S/CK34βE12/p63 are more helpful for diagnosis of prostate carcinoma,and it is important for diagnosis of benign and malignant lesions of the prostate.In the molecular range of 2-20k,there are 5 significant different peaks between PC and BPH(p<0.05).molecular 4624Da,1372Da,4346Da,3397Da are higher,and molecular 7764Da is lower in PC.Conclusions 3-antibodies cocktail staining play a more role of the detection rate of prostatic carcinoma.Combination immunohistochemical method and SELDI protein chip array technology is important in early diagnosis of prostatic carcinoma.
