Objective: To study the effect of propofol on the apoptosis and expression of Bcl-2 and Bax in small intestinal cells after Ischemic Reperfusion injury in rats and explore its mechanismMethods: Twenty-four rats were divided randomly into three groups: each consisting of 8 rats: Sham-operated control group, Ischemia-reperfusion+NS group (I/R+NS) and Ischemia-reperfusion +propofol group (I/R+P). Ischemic Reperfusion models were made by occluded the superior menseneric artery for 60 minutes followed by 120 minutes reperfusion. S and I/R+NS group were pretreated with Natrii Soldium 10ml·kg-1·h-1, and I/R+P were pretreated with propofol 10 mg·kg-1h-1 . Immunohistochemical staining were detected the expression of Bcl-2 and Bax, TUNEL method measured apoptotic intestinal cells and calculated the apoptotic index.Results: 1 .Histological evaluation S group: Normal. IIR+NS group: Epithelial cell necrosis, Villi effaced,more neutrophils in lamina propria, glandular destruction. I/R+P group: Epithelial cell degenerative changed but intact, mild inceased of lymphocytes and plasma cells in lamina propria. 2. The average OD value of Bax was significantly higher in I/R+NS group than that in S group (0.303±0.087 vs 0.101±0.035, P < 0.01) , but that was significantly lower in I/R+P group than that in I/R+NS group (0.167±0.061 vs 0.303±0.087, P < 0.01), and that was lower than in S group (0.167±0.061 vs 0.101±0.035, P < 0.05) the average OD value of Bcl-2 was higher in I/R + P group than that in I/R + NS group (0.242±0.062 vs 0.092±0.017, P < 0.01); and that still higher in I/R + NS group than in S group (0.092±0.017 vs 0.057±0.022, P < 0.05) . The Bax and Bcl-2 protein were located at cellular plasm, the positive cells had been stained yellow or brown. 3. The apoptotic index- in IIR+NS group was significantly higher than that in other two groups (0.449±0.019 vs 0.138±. 0.049; 0.449±0.019 vs 0.029±. 0.119, P<0.01), and that in I/R+P group was higher than that in S group(0.299±0.119 vs 0.138±0.049, P<0.05).The positive cells stained by TUNEL assay had brown cellular nuclei with blue cellular plasm.Conclusion: Propofol can reduce the apoptosis of intestinal mucosal epithelial cells during ischemia-reperfusion by accelerating the expression of bcl-2 gene and restraining the expression of bax gene, so can alleviate the injury of ischemia-reperfusion.
