Objective: With a high-cholesterol diet and vitamin D3 lavaged to establish experimental atherosclerosis (AS) model, and then studied the role of Tongnaoxinmailing (TNXML) in the prevention and treatment of AS and explored its mechanism. Methods: Except the control group, all the groups were lavaged with vitamin D3 three days, with a total dose of 500,000 IU/Kg; the control group was lavaged with equal volume of saline. Four weeks later, total cholesterol level checks of the serum and pathomorphology checks of rats were carried out. Compared the high-cholesterol diet groups with the control group, when there was an obvious difference, the treatment group were started to be lavaged with TNXML. Continuous administration was lasted for 8 weeks.After 12 weeks, biochemistry of the serum and morphologic changes of aortic pathology were observed. Results: (1) rat model of atherosclerosis was successfully established. (2) TNXML significantly lowered the levels of serum, TC, TG, LDL-C. Meanwhile it increased the levels of serum, HDL-C. (3) TNXML obviously enhanced the activity of SOD, cut down the content of MDA, and protected the vascular endothelium. (4)The examination in histomorphology showed TNXML could protect aortic EC of rats, prevent from forming elastic fibers and proliferation of smooth muscle.Conclusion: Tongnaoxinmailing can effectively improve the state of high blood cholesterol, inhibit lipid peroxidation, and protect vascular endothelial cells. Ultimately it can inhibit the formation of AS.
