BackgroundFibroblast activation protein (FAPa) was found by foreign scholars Rettig WJ in1986, He found a kind of membrane glycoprotein, was originally called F19antigen, It existed on activated fibroblast cell membranes, In normal adult fibroblasts usually no express or express level is very low. This suggested that it’s a marker on the surface of the fibroblasts, it plays a certain role in control of fibroblast growth epithelial tumors and embryonic development, and the process of wound repairing.During this thirty years, both at home and abroad about FAPa research work carried out gradually. Most concentrated on the study of tissue fibrosis and epithelial tumor.s A lot of research reports that FAPa plays an important role in the process of epithelial tumors. It’s mechanism has the following several possibilities. First, It’s functions of enzymes, directly dissolved extracellular matrix to assist in tumor cell invasion and participate in the reshaping of the tumor tissue; Second, dissociated and matrix proteins combined with growth factors, make it plays a role of the growth; Third, it involved in the generation of tumor micro vascular. FAPα has become a new cancer treatment targets and a hot spot of tumor clinical basic research, related research work is orderly carried out and the deficiency is still in the continuous improvement. As FAPa high expression of another part, both at home and abroad about the FAPa in wound repairing and reconstruction have rarely reported. It has already clear that FAPa existed in the tissue of wound and scar tissues, but it’s physiological role in the process of wound repairing and regulation mechanism not seen system researched and clarified.Objectives1. To observe the expression characteristics of FAPa in deep second scald wound repairing progress.2. To observe the expression characteristics of FAPa in pathological scar tissues.3. Discuss it’s significance in the process of scald wound repairing.Methods1. Forty eight health adult SPF Wistar rats were purchased from Southern medical university laboratory animal center, Male and female unlimited, weight of200~220g. According to the different phases of wound repairing were randomly divided into6h,12h and Id,3d,7d,14d,21d seven groups after scalded, each group of the sample size is6, and another6normal rats as the control group.2. Wistar rats with scalded model design reference literature,0.3%pentobarbital sodium (1ml/kg) after intraperitoneal injection of anesthesia, at room temperature, with90℃hot water scald rats skin for15s, Cause3.0cm diameter circular deep second degree scalded wound,(Pathological verification results for Ⅱ deep degree scald wounds). After injured each rat was raised in separate cage, the temperature is25℃, and daily routine dressing change by burns. Rats were executed and drawn at reservation time after injured. Take wounds central part1.0cm×1.0cm full-thickness skin tissue,1/2fixed in4%neutral paraformaldehyde, used in the immunohistochemical experiments, and the other1/2organization at-80℃refrigerator, used in western blotting test. Another6rats were directly executed after hair removal, take the same parts and size of skin tissue for control group.3. Six cases of pathological scar in limbs were selected from southern hospital which caused by deep second degree scalded.(each half of men and women, aged18to60, formation time is10-14months), Diagnosis depends on the medical history and pathological examination, pathological changes without any treatment, the local no burst, patients without systemic disease, at the same time take a few normal skin tissue around pathological scar for control group.4. Immunohistochemistry and western blotting technology were used to observe FAPa expression characteristics in tissues.5. Data processed by SPSS13.0software, measurement data with mean+/-standard deviation, according to the experimental data line Levene test, variance is consistent, the different time points in the process of wound repairing in rats FAPa gray value variance analysis, multiple comparison using SNK-q test. For scar tissue FAPα gray value line of the paired t test. α=0.05, the test of significance level when P<0.05, the difference was statistically significant.Results1. Immunohistochemical technique measure FAPa positive fibroblasts are dyed brown with DAB around the blood vessels and the newborn blood vessels of the wound.2. Western blotting technique measure FAPa gray value in skin tissues of rats is31.67+/-3.20,84.67+/-8.71,106.00+/-11.75,118.33+/-13.44,117.33+/-8.62,140.33+/-15.92,106.17+/-15.77,79.83+/-9.30in control group and experimental group. The gray value of different time points statistically significant difference with the control group.12h and1d,1d and3d FAPa of gray value there was no statistically significant difference (P>0.05), more than compared with the adjacent time point at each time point difference had statistical significance (P<0.05).3. Adult normal skin tissues FAPa gray value is18.00+/-7.87, pathologic scar tissues FAPα gray value is47.17+/-10.09, paired t test analysis results, the difference was statistically significant (P<0.05). Conclusion1. FAPa expressed in normal rat skin tissue, it gradually increased on6h after be scalded and reach its peak on7d, then gradually reduced, to21d after scalded FAP a expression is still have higher levels of expression.2. FAPaexpressed in normal adult skin tissue and pathological scar tissue, the expression level is higher than normal skin in pathological scar tissue.3. There is a close relationship between FAP a expression and activity characteristics of fibroblasts. It suggests that FAP a plays an important role in scald wound repairing process.
