Objective To observe the therapeutic effects of TACE combined withimmune therapy on transplanted liver neoplasm in Rabbits. Materials and MethodsThirty New Zealand White rabbits bearing transplanted VX2 liver cancer wererandomly grouped (6 rabbits each group) and treated by normal saline as control (G1),or TACE alone (G2), or TACE +injection of IL-2 and IFN-a via hepatic artery (G3),or TACE +injection of IL-2 and IFN-a via splenic artery (G4),or TACE combinedwith lipiodol emulsion+ IL-2 and IFN-a (G5).(TACE meant administration of THPlmg/kg +DDP 2mg/kg+lipiodol 0.2ml/kg, injection meant IL-2 100, OOOU/rabbitIFN- a 300, OOOU/rabbit, control group was injected into saline alone).Thetherapeutic effects were observed including analysis of changes of tumors'volume,eproportion of tumors'necrosis and the expression of PCNA LI in tumors'tissue. Thedistribution of T-lymphocytes in rabbits' peripheral blood were also detected withimmunofluorescence. Results①The 7th day after reatment,the tumor growthrabbit(GR) of G5 was significantly lower than that of G1~G4(P<0.05), and theG3,G4 lower than G2.The tumor growth inhibition rabbit of 5 was significantlyhigher than G2~G4(P<0.05).②The necrosis of G5 was significantly more extensivethan those of G1~G5(P<0.05). However, There was no significance of differencebetween the extent of necrosis of G3, G4 and G2 (P>0.05).③CD3~+,CD4~+T-lymphocytes in peripheral blood and CD4~+/CD8~+ proportion of G2 weresignificantly lower than those of G1(P<0.05), whereas those of G3 and G4 weresignificantly higher than those of G1(P<0.05), and those of G5 were significantlyhigher than those of G2(P<0.01) and G3,G4(P<0.05).④PCNA LI of G3, G4,G5was significantly lower than G1,G2(P<0.01).There was no significance of differencebetween G1 and G2. PCNA LI of G5 was significantly lower than G3, G4 P<0.05).Conclusion①It was obvious that an experimental therapeutic effect of TACE combined with lipiodol emulsion+ IL-2 and IFN-a on hepatic carcinoma. And it wasmore effective than administration of TACE alone or TACE +injection of IL-2 andIFN- a by hepatic artery or TACE +injection of IL-2 and IFN-a via splenic artery ininhibiting tumor growth.②TACE was one of the major cause leading to tumor'snecrosis, but it couldn't inhibit the proliferating activity of tumor cells. However,TACE combined with lipiodol emulsion+ IL-2 and IFN-a could take itevidently.③TACE could cause the descent of cellular immunologic function, whichshowed the disorder of T-lymphocytes population. TACE combined with lipiodolemulsion+ IL-2 and IFN-a could improve the immunologic function. In a word, itmay be an effective approach of hepatic carcinoma for TACE combined with lipiodolemulsion+ IL-2 and IFN-a. The author's study provides an experimental basis forclinical trial.
