Objective The present study was to investigate and confirm whether thepolypeptides from chlamys farreri(PCF)has the neuroprotective effect in anoxiccultured natal hippocampal neuronal cells. The ischemic cerebral diseases is thecommon diseases in clinic, The epidemical researches have shown that ischemiccerebral diseases, cancer and cardiopathy have becomed one of the three mostimportant causes of death. 70% of the survivors have different deformity, which bringabout heavy burden to family and society. The classic theory believes: The CentralNervous System can not recover once injured, but with developing of the science andtechnology and Molecular Biology, many scholars have affirmed that the CentralNervous System can also recover once was given proper conditions. That makespeople to be aware that the prevention and therapy to ischemic cerebral diseases isworkable. The present study emphasized on: The protective effects and mechanism ofthe PCF on hippocampal neuronal cells against anoxic.Methods Neurons in hoppocampal region of 1-3d newborned Wister rats werecultured by cytosinc arabinoside (Ara-c)-supplementing method, The neurons of the10-14d were cultured for 4h with 95%N2+5%CO2.Different dosage of PCF was givenafter anoxic for 4h to prevented therapy, consrasting with vitamin C. The activities ofthe neurons was tested by trypan blue method. The concentration of cellular freecalcium in the cells was tested by Fluorescence spectrophotometer(Fura-2). Themorphological changes was observed by Giemsa. The apoptosis rate and death rate ofhippocampal neuronal was tested by Hoechst.The contents of MDA and the activitiesof SOD in the supernatants of cells were determined by bicchemical methods.Results PCF significantly increased the activities of hippocampal neuronal cells,decreased the apoptosis rate and the death rate, decreased the concentration of cellularfree calcium in the cells, increased the activities of SOD and decreased the contents ofMDA in the supernatants of cells.Conclusions PCF has the neuroprotective effect on hippocampal neuronal cellsagainst anoxic. The mechanism of this effects of PCF may be mediated by decreasing the concentration of cellular free calcium in the cells, inhibiting the apoptosis of thecells, increasing the activities of antioxidative enzymes, eliminating the free radicalsto inhibit the oxidative damages caused by anoxic.
