Objective:To analysis the transcriptional expression of Zonula Occluden-1gene and the protein expression in oral verrucous carcinoma (OVC) and oral squamous cell carcinoma (OSCC),inquire into ZO-1related epithelial-mesenchymal transition in the development of oral cancer.Analysis the relationship between ZO-1expression with the clinic and pathology subtypes of oral cancer.Methods:The transcriptional expression of ZO-1was detected by Real-time PCR method,tissue Samples were obtained from cases of OSCC (n=10) and OVC (n=10) and normal oral mucosa epithelial specimens (n=10).The ZO-1protein expression is detected by immunohistochemistry, Paraffin specimens were obtained from cases of OSCC(n=20)and OVC(n=15)and normal oral mucosa epithelial(n=15).Results:1.The transcriptional expressions of ZO-1mRNA in OSCC and OVC were lower than normal oral mucosa tissue(p<0.05). The ZO-1 transcriptional level in OSCC was significantly lower than that in OVC(p<0.05).2.Compared between OVC,OSCC and normal epithelial group,The protein expression in OVC and OSCC are significant lower than normal mucosa epithelial. And there is no significant difference between exogenic OVC and high differentiated OSCC in protein expression.The expression in exogenic OVC is significantly higher than cyst-infiltrating type of OVC and the low differentiated OSCC,and all significantly lower than normal epithelial. The expression in cyst-infiltrating type of OVC has significant difference with low differentiated OSCC,and all lower than normal epithelial.The expression in high differentiated OSCC is significantly higher than the low differentiated OSCC, and all significantly lower than normal epithelial. The expression in invasive area of OSCC metastasis group is significant different with the invasive margin in non-metastasis group.3.The protein expression located in apical area of epithelium in normal mucosa. In OVC,the protein intensively expressed in apical exogenic area, but with a descend towards push-border. In high differentiated OSCC,ZO-1express strong in carcinoma nests, but present a diffuse distribution in low differentiated OSCC.ZO-1expression is positive in lymph node metastasis lesion, negative expressed in normal lymph node. Conclusions:1. As a tight junction protein, the expression of ZO-1protein and transcription level of mRNA is changed during the development and progression of oral cancer. It is functioned as a regulator of proliferation and differentiation and might play a role in cancer invasive and metastasis.2. The different characteristic expression in different types of OVC and OSCC, and the protein expression is related to differentiation, which could provide experiment evidence in explain the difference of pathological feature,biology behavior and clinic prognosis.3. ZO-1as an important protein in epithelium mesenchymal transition, its functioned as tight junction regulator could play a role in oral cancer invasive and metastasis.
