Study On The Efficacy And Mechanism Of Yiguanjian In Treatment Of Premature Ovarian Failure Rat Model

Purpose:With the help of premature ovarian failure rat model, this topic observes the efficacy and mechanism of action whose we use yiguanjian treating the rat through the angle of combining traditional Chinese and Western medicine, and then we preliminarily discuss the influence of yiguanjian on ovarian fuction.Material and method:Healthy female Wistar rat, weighing190-230g. Normal feeding after1week,The60rats who were weight and detected estrogen level by tail vein blood were incorporated experiment at estrus. These rats were random divided into3groups:normal control group, model group, yiguanjian group. The experiment take Tripterygium wilfordii as premature ovarian failure model drug, model group and yiguanjian group dose rats with Tripterygium wilfordii according to0.6mg/100mg (weight) for9days, and gavage once a day; The normal control group was given the same dose of physiological saline for9days once a day. After modeling, all rats were detected estrogen level by tail vein blood. After the tenth day, The normal control group and model group was given the same dose of physiological saline for15days once a day, and yiguanjian group dose rats with yiguanjian for15days once a day. After the drug, the rats were weight and drown blood in tail vein within24hours, then we take out ovary by the internal fixation methods. We weigh the rat body weight, and obverse rat’s weight changes and behavior state changes before and after the experiment. After the rats were taken out blood, thought determining the content of follicle stimulating hormone (FSH) and luteinizing hormone (LH) and estradiol (E2) by immunological method, we belive that the model is successful:and then we make the ovary paraffin sections and HE staining to observe the change of ovary structure.Results: 1. The body weight and behavior results show:Compared to before the experiment, the rats of model group weight loss, and there was statistical significance. The rats are animal in good condition, quick, smooth coat, and normal in eating food in Control group. After the experiment, The rats of Control group are No obvious abnormality,but Most of the model rats show lazy, don’t love sports, unresponsive, shaggy, brilliance, Color haggard, reduce appetite with the passage of molding time, after given decoction treatment, the rats of yiguanjian group show:body weight increased obviously.activity increased gradually, diet increased, then they gradually returned to normal level.2. The content change of E2in serum results show:Compared with the normal group, the content of E2in serum decreased significantly(p<0.01), and there is obvious statistical significance in the model group, and show the model is successful. And after dosing rats with yiguanjian in the yiguanjian group, the content of E; increased significantly in comparison with the model group (p<0.01), and reached the same level of the normal group.3. The content change of FSH in serum results show:Compared with the normal group, the content of FSH in serum increased significantly(p<0.01), and there is obvious statistical significance in the model group, and show the model is successful. And after dosing rats with yiguanjian in the yiguanjian group,the content of FSH decreased significantly in comparison with the model group (p<0.01), and reached the same level of the normal group.4. The content change of LH in serum results show:Compared with the normal group,the content of LH in serum increased significantly (p<0.01), and there is obvious statistical significance in the model group, and show the model is successful. And after dosing rats with yiguanjian in the yiguanjian group, the content of LH decreased significantly in comparison with the model group (p<0.01),and reached the same level of the normal group.Conclusion:1. Body weight of premature ovarian failure’rat show clear downward trend, behavioral states of them have the obvious change-lazy, don’t love sports, unresponsive, shaggy, brilliance, Color haggard,reduce appetite and other changes. after given decoction treatment,the rats of yiguanjian group show:the body weight increased obviously and the state improved2. Traditional Chinese medicine yiguanjian has good therapeutic effect on premature ovarian failure rat model.3. Yiguanjian can effectively improve the overian function of the premature ovarian failure rat model.
