Expression Of Survivin And High-risk Human Papillomavirus DNA And Their Relationship In Laryngeal Squamous Cell Carcinoma

Purpose To investigate the expression of Survivin and HPV16/18 DNA,and their co-relationship in laryngeal squamous cell carcinoma(LSCC) by immunohistochemistry and fluorescence quantitative polymerase chase reaction (FQ-PCR)experiment.Methods 48 samples of laryngeal squamous cell carcinoma were collected.The samples were cut off by surgical operation from otorhinolaryngology-head and neck surgery section of the people's hospital of hunan province during the period 2001 to 2006. 20 samples normal squamous epidermis near cancer and 20 samples laryngeal polypi were collected for contract.The patients' age, sex, differentiation, lymph node transfer and TMN stage were recorded. All samples were re-cuted to diagnose. Survivin was done by SP immunohistochemical staining and the results of immunohistochemistry experiment were determined by intensity of staining and the number of positive cells. The expression of HPV16/18 DNA was done by FQ-PCR. Analyse the expression of Survivin in LSCC,normal squamous epidermis near cancer and laryngeal polypi.Analyse the relationgship between the expression of Survivin and HPV16/18 DNA with the clinical pathology characteristics.Analyse the relationship of the expression of Survivin and HPV16/18 DNA.Result1 The expression of Survivin in LSCC was up-regulated,and the expression had significant difference (P<0.05) between LSCC and normal squamous epidermis near cancer or laryngeal polypi. There was not significant correlation between the expression of Survivin and the age of patient, type of pathology,but there was significant correlation between the expression of Survivin with the lymph node transfer and tumor TMN stage.2 The expression of HPV16/18 DNA in LSCC was up-regulated,and the expression had significant difference (P<0.05) between LSCC with normal squamous epidermis near cancer,laryngeal polypi. There was not significant correlation between the expression of HPV16/18 DNA with type of pathology,the tumor TMN stage and lymph node transfer.3 The expression of Survivin and HPV16/18 DNA has positive correlation (r=0.801;P<0.01).There was a cooperation between the expression of Survivin and HPV16/18 DNA in LSCC. Conclusion1 The up-regulated expression of Survivin in LSCC may be an important function in genesis and progress of LSCC.2 The infection HPV16/18 may have a role of spring in LSCC.3 There is a cooperation between Survivin and HPV16/18DNA in LSCC.
