Objective:To investigate the role and the expression of c-jun in the pathogenesis of colitis in rats by using Uyghur's medicine Kui Jiean and to look for the mechanism of Kui Jiean in rat's colitis by using RNA interference.Methods:1.Wister rats were randomly divided into five groups:normal group,model group,NS group,5-ASA therapeutic group and Kui Jiean therapeutic group;rat colitis model was induced by DNCB combined with acetic acid,the expression of c-jun in the rat's intestin were detectde by semi-quantatitive reverse transcription-polymerase chain raction(RT- PCR)and Western blot technique. 2.To contruct pcDNA3.1/CT-GFP-TOPO/c-jun and the siRNA expression vector of rat c-jun gene.Results:1.At the level of mRNA:the expression of c-jun between five groups have no statistic signifinance(P=0.071).at the protein level:the expression of c-Jun in modle group was higher than that of normal group(P<0.05);the expression of c-Jun in Kui Jiean therapeutic group has statistic signifinance compared with model group and NS group.2.Successfully construction of pcDNA3.1/CT-GFP-TOPO/ c-jun and the siRNA expression vector of rat c-jun gene.Conclusion:1.c-jun may participate in the occurrence and development of ulcerative colitis and Uyghur's medicine Kui Jiean may cure the Ulcerative Colitis in rat by deregulating the expression of c-jun.2.The successful construction of pcDNA3.1/CT- GFP-TOPO/c-jun and the siRNA expression vector of rat c-jun gene lays the basis for RNA interference technique,which will enable us to develop an approach to study the mechanism and new therapy method.
